化瘀通孕方对输卵管炎性阻塞性不孕症大鼠ICAM-1表达及血流变学的影响 被引量:8

摘要 目的:研究化瘀通孕方对输卵管炎性阻塞性不孕症大鼠输卵管细胞间黏附分子-1(ICAM-1)表达及血流变学的影响,从分子生物学角度探讨化瘀通孕方的作用机制。方法:SPF级Wistar雌性大鼠70只,随机选取60只大鼠采用混合菌法制作输卵管炎性不孕症大鼠模型,剩余10只作为空白组。选择造模成功的大鼠50只,随机分为模型组、西药组、化瘀通孕方低、中、高各剂量组,各10只。化瘀通孕方低、中、高各剂量组分别按6.09、12.18、24.36 g/(kg·d)灌胃化瘀通孕方,西药组灌胃头孢呋辛酯片52.5 mg/(kg·d)和甲硝唑片63 mg/(kg·d),空白组和模型组予等容量生理盐水灌胃。检测大鼠血液流变学指标和输卵管组织ICAM-1蛋白的表达。结果:空白组大鼠输卵管不表达ICAM-1。与空白组比较,模型组大鼠全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著升高(P<0.01),说明造模成功。与模型组比较,西药组和化瘀通孕方低、中、高各剂量组大鼠输卵管ICAM-1蛋白表达累积面积和平均光密度均显著减小,全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著降低,差异均有统计学意义(P<0.01)。与西药组比较,化瘀通孕方低剂量组大鼠输卵管ICAM-1蛋白表达累积面积较高;化瘀通孕方中、高剂量组大鼠输卵管ICAM-1蛋白表达累积面积、大鼠全血黏度高切和低切、红细胞刚性指数、红细胞压积均显著降低,差异均有统计学意义(P<0.05)。与化瘀通孕方高剂量组比较,化瘀通孕方低、中剂量组大鼠输卵管ICAM-1蛋白表达累积面积和平均光密度均显著较高,全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著较高,差异均有统计学意义(P<0.05)。结论:化瘀通孕方能明显改善大鼠血液流变学指标,并可以降低输卵管ICAM-1蛋白的表达,为临床治疗输卵管炎性阻塞性不孕症提供可靠的实验依据。 Objective:To study the effect of Huayu Tongyun prescription on intercellular adhesion molecule-1(ICAM-1)expression and hemorheology of rats with infertility due to inflammatory obstruction of fallopian tube. Methods :There are 70Wistar female rats of SPF grade and selected 60 rats of them randomly to build models of infertility due to inflammatory obstruction of fallopian tube with mixed culture method,the other ten rats were selected as the blank group. Selected 50 successful model rats and divided them into the model group,western medicine group,and low,middle and high dose of Huayu Tongyun prescription groups randomly, 10 cases in each. The low, middle and high dose of Huayu Tongyun prescription groups were given 6.09, 12.18, 24.36 g/(kg·d) of Huayu Tongyun prescription respectively by gavage. The western medicine group was given Cefuroxime axetil tablets of 52.5 mg/(kg·d) and Metronidazole tablets of 63 mg/(kg·d) by gavage, and the blank group and the model group were given isovolumetric saline solution. Detected ICAM-1 protein expression of ovarian tissues and hemorheology indexes of rats. Results:There was no expression of ICAM-1 of ovarian tissues in the rats of the blank group. Comparing with those of the blank group,whole blood viscosity in high shear rate and in low shear rate,erythrocyte aggregation and rigidity index and hematocrit of the model group were all increased significant(P〈0.01),which suggested the success of modeling. Comparing with those of the model group,cumulative area and average optical density of ICAM-1 protein expression in ovarian tissues of the western medicine group and the low,middle and high dose of Huayu Tongyun prescription group were all reduced significantly,and the whole blood viscosity in high shear rate and in low shear rate,erythrocyte aggregation and rigidity index and hematocrit were also reduced significantly,the differences being significant(P〈0.01). Comparing with those in the western medicine group,cumulative area of oviduct ICAM-1 protein expression in the low dose of Huayu Tongyun prescription groups was higher. Cumulative area of oviduct ICAM-1 protein expression,and the whole blood viscosity in high shear rate and in low shear rate,erythrocyte aggregation and rigidity index and hematocrit in the middle and high dose of Huayu Tongyun prescription groups were all reduced significantly(P〈0.05).Cumulative area and average optical density of ICAM-1 protein expression in ovarian tissues in the low and middle dose of Huayu Tongyun prescription groups were significantly higher than those in high dose of Huayu Tongyun prescription group,and the whole blood viscosity in high shear rate and in low shear rate, erythrocyte aggregation and rigidity index and hematocrit were also significantly higher than those in high dose of Huayu Tongyun prescription group(P〈0.05). Conclusion:Huayu Tongyun prescription can improve hemorheology indexes of rats significantly,and reduce the oviduct ICAM-1 protein expression,which can provide reliable experiment basis for treating infertility due to inflammatory obstruction of fallopian tube.
