miR-423、miR-205和miR-1307单核苷酸多态性与子宫颈癌的相关性研究 被引量:1

摘要 目的探讨mi R-423、mi R-205和mi R-1307单核苷酸多态性与宫颈癌的相关性。方法以病例-对照研究的方法收集104例宫颈癌及186例骨折、高血压等同期非肿瘤患者为对照,采用PCR-限制性酶切片段长度多态性技术对mi R-423 rs6505162位点进行基因型分析;采用PCR和多重连接酶检测反应技术对mi R-205 rs3842530和mi R-1307 rs7911488基因多态性进行分析。结果 rs6505162 CC、rs6505162 AC和rs6505162AA基因型在宫颈癌组中分布为64.4%、31.7%和3.8%,对照组分布为66.1%、30.1%和3.8%;rs3842530-/-、-/insert和inser/inser基因型在宫颈癌组中分布为56.7%、34.6%和8.7%,对照组分布为52.7%、36.0%和7.0%;rs7911488 AA、AG和GG基因型在宫颈癌组中分布为51.9%、39.4%和8.7%,对照组分布为45.1%、45.1%和9.7%。各位点基因型分布比较,差异均无统计学意义(P>0.05)。进一步按年龄分层分析显示,≤50岁和>50岁不同年龄组各基因型分布在对照组和宫颈癌组中比较,差异无统计学意义(P>0.05)。结论 mi R-423 rs6505162、mi R-205 rs3842530和mi R-1307 rs7911488基因多态性可能与宫颈癌发生无关。 Objective To investigate the relationship between gene polymorphism of miR-423,miR-205 and miR-1 307 with cervical cancer risk. Methods A case-control study was carried out including 104 patients with cervical cancer and 186 controls such as fracture and hypertension during the same period. Mi R-423 rs6505162 polymorphism was examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) assay. The gene polymorphism of miR-205 rs3842530 and miR-1307 rs7911488 were detected by multiple PCR and multiple LDR method. The association between the genotypes and cervical cancer was analyzed by Chi-square test. Results The genotype of rs6505162 were 64.4% for CC, 31.7% for AC, 3.8% for AA in cases and 66.1% for CC, 30.1% for AC, 3.8% for AA in controls. The genotype of rs3842530 were 56.7% for-/-, 34.6%for-/insert, 8.7%for inser/inser in cases and 52.7% for-/-, 36.0% for-/insert, 7.0% for inser/inser in controls. The genotype of rs7911488 were 51.9% for AA, 39.4% for AG, 8.7% for GG in cases and 45.1% for AA, 45.1%for AG, 9.7% for GG in controls. The genotype distribution of miR-423, miR-205 and miR-1307 polymorphism in the cases and the controls had no statistically significant difference(P〉0.05). Further agestratified analysis showed that these three polymorphism genotype frequencies distribution had no statistical significance between ≤ 50 years old and 〉50 years old in the cases and the controls(P〉0.05). Conclusion There are no association between gene polymorphism of miR-423 rs6505162, miR-205 rs3842530 and miR-1307 rs7911488 and cervical cancer.
