摘要
目的应用实时荧光定量PCR技术对2015年陕西炭疽疫情快速鉴定和种群分析。方法对甘泉县炭疽病例水泡液或病灶涂抹物及血液标本进行细菌分离,同时提取核酸进行荧光定量PCR试验。结果从1例病灶涂抹物中分离到1株炭疽芽胞杆菌。在19例病例中检出16例核酸阳性,疫情相关菌株种群确定为A.Br.001/002群。结论炭疽杆菌实时荧光定量PCR可用于炭疽疫情迅速判定,确定病例菌株种群,可克服传统病原培养分离阳性率低,血清学诊断时间长的缺点。
[Objective]To carry out the rapid identification of anthrax epidemic situation and molecular typing in Shaanxi in 2015 by real-time fluorescent quantitative PCR. [Methods]The bacteria were isolated from blister exudate,lesion smear and blood specimens of anthrax cases in Ganquan County. Meanwhile,the nucleic acid was extracted for fluorescent quantitative PCR.[Results]1 strain of Bacillus anthracis was isolated from the lesion smear of one case. Among 16 cases,the positive nucleic acid was detected in 16 cases,and the epidemic strain was identified as A.Br. 001/002 group. [Conclusion]Real-time fluorescent quantitative PCR can be used for the rapid determination of the anthrax epidemic situation,and determine the groups of the strains.It can overcome the disadvantages of low positive rate and long serological diagnosis time of traditional pathogenic culture method.
