摘要
目的:观察黄芪制首乌汤剂对阿霉素大鼠肾病模型生化指标及肾组织的影响,探讨复方及其君药对肾功能的保护作用。方法:建立阿霉素肾病(ADRN)模型,造模成功后将35只大鼠随机分为5组,即空白组、ADRN模型组(模型组)、益肾通络方治疗组(复方组)、复方减君药治疗组(去君药组)、黄芪+制首乌治疗组(君药组),每组7只。治疗组每天分别灌胃益肾通络方复方汤剂(4.58g/kg)、复方去君药汤剂(3.44g/kg)、黄芪+制首乌汤剂(1.14g/kg),空白组和模型组灌胃等量蒸馏水,连续3周后检测大鼠的生化指标和肾组织病理变化。结果:模型组、复方组、去君药组、君药组24h uPro用药前后比较,差异均有统计学意义(P<0.01)。用药后,复方组、去君药组、君药组24h uPro、ALB、Scr水平与模型组比较,差异均有统计学意义(P<0.01,P<0.05);去君药组ALB、Scr、BUN水平及君药组Scr、BUN水平与复方组比较,差异均有统计学意义(P<0.01,P<0.05);君药组ALB、Scr、BUN水平与去君药组比较,差异均有统计学意义(P<0.01)。各治疗组大鼠病理学检查均有一定程度恢复。结论:中药复方及其君药对模型大鼠肾功能有良好的保护作用,君药在复方中不可或缺。
Objective: To observe the effects of astragalus blood biochemical parameters and renal histopathological (ADRN) , and to investigate the protective effect of kidney and Radix PolygoniMuhiflori decoction on the changes in rats with adriamycin nephropathy - nourishing and collateral - dredging compound prescription and its monarch drug on their renal function. Methods:An ADRN model was established, and a total of 35 rats were randomly and equally divided into 5 groups: blank group, ADRN model group (model group) , kidney - nourishing and collateral - dredging compound prescription group ( compound group) , kidney - nourishing and collateral - dredging compound prescription without monarch drugs group ( no monarch drug group), and astragalus plus Radix PolygoniMuhiflori group (monarch drug group), with 7 rats in each group. The three treatment groups were given kidney - nourishing and collateral - dredging compound decoction(4.58g/kg) , kidney - nourishing and collateral - dredging compound decoction without monarch drugs (3.44g/kg), and astragalus plus Radix PolygoniMultiflori decoction( 1. 14g/kg), respectively, by gavage, while the blank group and the model group were given the same volume of distilled water by gavage. After 3 weeks of administration, the blood biochemical parameters and renal histopathological changes of rats in each group were observed. Results:The model group, compound group, no monarch drug group, and monarch drug group had significant changes in 24h urinary protein excretion (24h uPro) after treatment (P 〈 0.01 ). After treatment, there were significant differences between the model group and the compound group, no monarch drug group, and monarch drug group inlevels of 24h uPro, albumin ( Alb), and serumcreatinine (Scr) ( P 〈0.01 ,P 〈 0.05 ) ; there were significant differences in levels of Alb, Scr, and blood urea nitrogen ( BUN ) between the no monarch drug group and the compound group, as well as significant differences in levels of Scr and BUN between the monarch drug group and the compound group (P 〈 0.01, P 〈 0.05 ) ;there were signifi- cant differences in levels of Alb, Scr, and BUN between the monarch drug group and the no monarch drug group ( P 〈 0.01 ). The three treatment groups showed improvements in renal function on histopathological examination. Conclusion:The kidney - nourishing and collateral - dredging compound prescription has a good protective effect on the renal function of rats with ADRN, and its monarch drugs are indispensable.
