卡介菌多糖核酸对CIK细胞活性的影响 被引量:2

The Impact of BCG-PSN on the Activity of CIK Cells

摘要 在CIK细胞的培养过程中加入卡介菌多糖核酸,研究其对CIK细胞体外增殖及杀瘤活性等方面的影响.CCK-8检测结果发现低浓度卡介菌多糖核酸处理的CIK细胞相比对照组(生理盐水),细胞增殖能力和杀瘤效果都显著上调.RT-qPCR结果显示卡介菌多糖核酸处理后的CIK细胞毒活性相关因子(Granzyme B,IFN-γ,TNF-α和TNF-β)mRNA的表达相比对照组都有提高.这些结果可为培养具有更高效杀瘤活性又能大量扩增的CIK细胞提供新思路. The BCG polysaccharide and nucleic acid (BCG-PSN) was added to CIK ceils to study CIK cells' responses on the proliferation and tumoricidal activity in vitro. CCK-8 results showed that the CIK cells treated with low dose BCG-PSN, compared with control group (normal saline), were significantly enhanced on the proliferation and tumoricidal activity. RT-qPCR results revealed that the expression of CIK Granzyme B, IFN-T, TNF-α, TNF- β mRNA of BCG-PSN group was higher than that of the control group. These results lay the foundation for how to cultivate a large number of CIK cells with high tumoricidal activity, and provide new ideas for the therapy of anti- tumor adoptive immunity.
