摘要
胰岛素样生长因子(insuline-like growth factors,IGFs)是一类具有胰岛素样代谢和促进有丝分裂功能的多肽,包括IGF1和IGF2,对调节垂体生长激素合成和分泌起着重要的作用,同时还参与了细胞的增殖、生长、分化等过程.利用RT-PCR在mRNA水平上检测了IGF1/2基因在金鱼不同组织和不同胚胎发育时期的表达.结果表明,IGF1基因在肝脏中的表达量最高,在大脑、肌肉及精巢的表达量相对较低;IGF2基因在脾脏中的表达量最高,在肾脏、卵巢及精巢的表达量相对较低.IGF1/2基因在不同发育时期基本上呈现由低到高的趋势.研究表明,IGF1/2基因在金鱼的不同组织和胚胎发育过程中可能具有重要的作用,但其具体机制还有待于进一步研究.
Insulin-like growth factors ( IGFs), including IGF1 and IGF2, are a class of polypeptides with insulin-like metabolic and mitogenic functions. It not only plays an important role in regulating pituitary hormone synthesis and secretion, but also involved in cell proliferation and growth and differentiation processes. In this work, RT-PCR was used to examine the expression of IGF1/2 gene in different tissues and periods of embryonic develop- ment at the mRNA level in goldfish. Our results reveal that the IGF1 gene was expressed the highest level in the liver and to a much less degree in the brain, muscle and testis of goldfish, whereas IGF2 was expressed the highest in the spleen but relatively low in the kidney, ovary and testis. The IGF1/2 genes were expressed differently at different developmental stages, with the pattern of "low to high" trend. These studies suggest that IGF1/2 genes should play important roles in embryonic development and in different tissues of goldfish. The exact mechanism remains to be further investigated.
