高效毛细管电泳法同时检测苹果中4种多酚类化合物 被引量:7

Simultaneous determination of 4 phenolic compounds in the apples by high performance capillary electrophoresis

摘要 利用高效毛细管电泳法(HPCE),对苹果中根皮苷,表儿茶素,芦丁,绿原酸4种多酚类化合物进行分离与测定。在最佳实验条件:缓冲溶液为10 mmol/L Na_2B_4O_7-H_3BO_3,pH 9.2,检测波长280 nm,分离电压25 kV,测定了4种多酚类化合物的标准品,其线性范围分别为0.005~0.120,0.005~0.550,0.004~0.450,0.020~0.600 mg/mL,相关系数r在0.9977~0.9992之间,检出限分别为34.1,56.7,37.8,38.9μg/L(S/N=3)。日内及日间精密度和平均回收率范围分别为0.13%~4.5%和为96.7%~104.5%,相对标准偏差(RSD)≤3.7%(n=3)。方法可用于苹果中多种酚类物质的同时检测。 A method of high performance capillary electrophoresis (HPCE) was developed for the simultaneous separation and determination of ploridzine, ( - ) - epicatechin, rutin and chlorogenic acid in apples. The effects of different electrophoresis conditions were investigated. The running buffer was 10 mmol/L Na2B407 -H3BO3 at pH 9.2, and the detection wavelength was set at 280 nm and applied voltage at 25 kV. Under these optimal conditions, ploridzine; ( - ) - epicatechin; rutin and ehlorogenic acid could be fully separated from each other within 7 minutes. The linear ranges were 0. 005 to 0. 120, 0. 005 to 0. 550, 0. 004 to 0. 450 and 0. 020 to 0. 600 mg/mL, respectively, with the correlation coefficients between 0. 9977 and 0. 9992. The lowest detection limits were 34. 1,56. 7, 37.8,38.9 μg/mL, respectively ( S/N = 3 ). The relative standard deviations of migration time and peak area were between 0. 13% and 4. 5%. The average recoveries of spiked samples were in the range from 96. 7% to 104. 5 %, with a relative standard deviations (RSD) lower than 3.7% (n = 3 ). This method is rapid, simple, accurate and reliable for the simultaneous detection of phenolic compounds in apples.
