解毒化瘀颗粒影响急性肝衰竭大鼠肝细胞周期的实验研究

Experimental Study on Effect of Jiedu Huayu Granules on Hepatic Cell Cycle in Rats with Acute Liver Failure

目的:明确解毒化瘀颗粒对急性肝衰竭大鼠肝细胞有丝分裂指数和增殖指数的影响,为深入研究奠定基础。方法:以D-氨基半乳糖(D-Gal N)联合内毒素脂多糖(LPS)构建急性肝衰竭大鼠模型,在造模后6 h、24 h、48 h每组抽取6只大鼠采取标本,进行肝细胞有丝分裂指数和增殖指数检测。结果:急性肝衰竭大鼠经解毒化瘀颗粒干预治疗后,肝细胞在造模后24 h、48 h有丝分裂指数和增殖指数明显提高,与模型对照组比较差异具有统计学意义(P<0.05)。结论:解毒化瘀颗粒可促进急性肝衰竭大鼠肝细胞增殖与分裂,其机制与提高肝细胞在S期DNA复制有关。

Objective：To confirm the influence of Jiedu Huayu Granules on the mitotic index and proliferation index of liver cells in rats with acute liver failure and establish the basis for further research.Methods：Build a rat model of acute liver failure with D-galactosamine（D-Gal N） combined with endotoxin lipopolysaccharide（LPS）.The detection of cell mitosis index and proliferation index was at 6 h,24 h,48 h from 6 rats of each group after injecting D-Gal N and LPS.Results：After the intervention treatment of acute liver failure in rats by Jiedu Huayu Granules,mitosis index and proliferation index of liver cells in the model at 24 h,48h increased significantly.Compared with the model group,the difference was statistically significant（P〈0.05）.Conclusion：Jiedu Huayu Granules can promote the proliferation and division of hepatic cells in rats with acute liver failureand the mechanism is related to the increase of DNA replication in S phase.
