摘要
目的:探讨脾虚痰浊证巴马小型猪心肌组织葡萄糖代谢相关分子的变化特点。方法:普通级雄性巴马小型猪10只随机分为正常组及脾虚痰浊组,每组5头。采用疲劳过度联合高脂单笼饲养建立小型猪脾虚痰浊证模型。采用实时荧光定量PCR法检测心肌组织葡萄糖转运体4(glucose transporter 4,GLUT4)、磷酸果糖激酶1(Phosphofructokinase-1,PFK-1)、丙酮酸激酶(Pyruvate kinase2,PKM2)和己糖激酶(Hexokinase,HK)mRNA表达,应用Western blotting法检测心肌组织GLUT4、PFK1、PKM2和HK蛋白表达水平。结果:与正常组小型猪相比,脾虚痰浊组小型猪心肌组织GLUT4、PKM2、PFK1、HK mRNA表达水平显著下降(P<0.01),GLUT4、PKM2蛋白表达水平显著下降(P<0.05)。PFK1、HK蛋白表达水显著下降P<0.01)。结论:脾虚痰浊证小型猪心肌存在葡萄糖代谢异常,与葡萄糖转运及代谢相关分子表达呈下降趋势。
Objective:To investigate the changes of glucose metabolism related molecules in cardiac muscle of Bama minipigs with spleen deficiency and phlegm syndrome.Methods:Ten common grade Male Bama minipigs were randomly divided into normal group,spleen deficiency and phlegm group,5 pigs in each group.The model of spleen deficiency and phlegm syndrome was established by the combination of fatigue and high-fat feeding seperately.The expression levels of GLUT4,PFK1,PKM2 and Hexokinase mRNA were detected by real-time fluorescence quantitative PCR method and the expression levels of GLUT4,PFK1,PKM2 and HK protein in cardiac muscle tissue were detected by Western blotting method.Results:Compared with the normal group,in the myocardium of minipigs with the spleen deficiency and phlegm syndrome,the expression levels of GLUT4,PKM2,PFK1 and HK mRNA were significantly decreased(P〈0.01).The expression levels of GLUT4 and PKM2 protein were significantly decreased(P〈0.05).The expression levels of PFK1 and HK protein were significantly decreased(P〈0.01).Conclusion:Glucose metabolism disorder exists in cardiac muscle of Bama minipigs with spleen deficiency and phlegm syndrome and the expression of glucose transporter and transformation related molecules decreased.
