宁夏密点麻蜥对胃癌前病变模型大鼠P53及PUMA表达的影响

Impact of Ningxia Dense Point Hemp Lizard on P53 and PUMA Expressions on Precancerous Lesion of Gastric Carcinoma Model Rats

摘要 目的:研究宁夏密点麻蜥不同剂量对胃癌前病变(PLGC)模型大鼠胃黏膜细胞增殖、凋亡以及P53和PUMA水平的影响,探讨宁夏密点麻蜥对PLGC的干预机制。方法:60只SPF级雄性SD大鼠,随机分为空白对照组,模型组,宁夏密点麻蜥高、中、低剂量组,维霉素对照组,共6组,每组10只。除空白组外,余均以0.02 mmol/L的MNNG联合40%乙醇、饥饱失常等综合因素共24周制备PLGC大鼠模型。分别用宁夏密点麻蜥高、中、低剂量、维酶素连续治疗12周,观察PLGC模型大鼠胃黏膜病理组织学变化,应用免疫组化法分别观察给药前和停药后胃体组织P53和PUMA蛋白变化,并进行统计分析。结果:通过Image-Pro Plus 6.0图像分析系统对免疫组化图像进行光密度的测算,各模型组与空白对照组P53和PUMA水平比较,AOD值明显降低,P<0.05;各治疗组与模型组P53和PUMA水平比较,AOD值明显降低,P<0.05;宁夏密点麻蜥高、中、低剂量治疗组P53和PUMA阳性表达率低于维酶素治疗组,P<0.05,其中以高剂量组AOD值最小。结论:宁夏密点麻蜥能抑制凋亡相关蛋白P53、PUMA的表达,促进细胞凋亡,逆转PLGC模型大鼠胃黏膜病理改变,阻断癌变发展。 Objective:To study the Ningxia dense point hemp lizard different dosages on gastric mucosal cell proliferation,apoptosis and p53 and puma level influence of the precancerous lesions of gastric carcinoma(PLGC) model rats and explore the ningxia dense point hemp lizard on intervention mechanism of PLGC.Methods:Sixty SPF male SD rats were randomly divided into blank control group,model group,the Ningxia dense point hemp lizard high,medium,low dosages groups,Vitacoenzyme treatment group,10 rats in each group.Except for the blank group,prepare PLGC rats model with 0.02 mmol/L MNNG,combined with 40%ethanol,hunger and satiety disorders comprehensive factors for 24 weeks.Respectively with the Ningxia dense point hemp lizard high,medium and low dosages and Vitacoenzyme,continuous treatment 12 weeks.Observe gastric mucosa pathological changes and p53 and PUMA protein changes by immunohistochemical staining and statistical analysis.Results:By Image Pro Plus 6.0 image analysis system on immunohistochemical image optical density estimates,the model group with blank control group's p53 and PUMA level and AOD values decreased significantly,P〈0.05.Each treatment group and model group's p53 and PUMA level and AOD values decreased significantly,P〈0.05.The Ningxia dense point hemp lizard high,medium and low dosages treatment groups' PUMA and p53 positive expression rate were lower than those of Vitacoenzyme treatment group,P〈0.05.The high dosages group's AOD was minimum.Conclusion:The Ningxia dense point hemp lizard can inhibit the expression of apoptosis related proteins p53 and PUMA,promote cell apoptosis,reverse PLGC model rats gastric mucosa pathological changes and block the development of cancer.
