摘要
目的:利用PCR array技术检测脾虚痰浊动脉粥样硬化巴马小型猪冠脉应激反应与脂质转运和代谢相关基因变化,探讨脾虚痰浊冠状动脉粥样硬化发生的病理机制。方法:随机将10只健康的广西巴马小型猪分为正常组和模型组,每组5只,正常组给予普通饲料饲喂,模型组给予球囊拉伤术联合高脂饮食饲喂进行脾虚痰浊动脉粥样硬化模型复制,实验24周后,观察巴马小型猪行为学变化。呼吸机麻醉后取材,分离血清和冠状动脉,测定血清血脂、hs-CRP、IL-6水平,HE染色观察冠状动脉组织形态学变化,对模型进行评价。应用PCR array技术观察巴马小型猪冠脉应激反应与脂质转运和代谢相关基因变化。结果:造模24周后,巴马小型猪出现食少且不欲饮食、神疲乏力、倦怠等明显脾虚证候,与正常组比较,模型组血清血脂、hs-CRP、IL-6水平显著升高,HE染色结果表明,模型组小型猪血管内膜损伤严重,提示模型复制成功。PCR array结果显示,与正常组比较,与应激反应相关的25个基因中,上调11个,下调3个,与脂质转运和代谢相关的15个基因中,上调5个,下调5个。结论:脾虚痰浊巴马小型猪冠脉应激反应与脂质转运和代谢相关基因变化可能是脾虚痰浊冠状动脉硬化发生的病理机制。
Objective:To investigate the pathological mechanism of the coronary atherosclerosis with spleen deficiency and phlegm syndrome and detect the changes of response to stress,lipid transportation and metabolism related genes in coronary artery from spleen deficiency and phlegm syndrome Bama Miniature pigs by PCR array technology.Methods:Totally 10 Bama Miniature pigs were randomly divided into the control group and the model group,5 in each group.Common granular forage was fed to pigs in the control group.Pigs in the model group were treated with balloon injury and high-fat forage to establish the spleen deficiency and phlegm syndrome Bama miniature pig model.After 24 weeks of experiment,observe the change of Bama miniature pigs' behaviors.After anesthesia,separate the serum and coronary artery and test the levels of lipids,hs-CRP and IL-6 in serum.HE staining was used to observe the morphological changes of the coronary artery and evaluate the model.PCR array technology was used to test the change of genes related with response to stress,lipid transportation and metabolism in the coronary artery of Pigs.Results:24 weeks after modeling,eat less,do not want to diet,fatigue,lassitude and burnout symptoms with spleen deficiency and phlegm syndrome were appeared in the model group pigs.Compared with control group,serum levels of lipids,hs-CRP and IL-6 were higher in the model group.HE staining results showed that the vascular intima injury was serious of model group,suggesting the modeling success.PCR array results showed that compared with control group,among the 25 genes related with stress response,12 were up-regulated and 3 were down regulated.Among the 16 genes related with lipid transportation and metabolism,5 were up-regulated and 5 were down regulated.Conclusion:The changes of response to stress,lipid transportation and metabolism related genes in coronary artery may be the pathological mechanism of the coronary atherosclerosis.
