长毛风毛菊醇提物对ANIT所致小鼠肝损伤的保肝作用研究 被引量:4

摘要 目的:观察长毛风毛菊醇提物对α-异硫氰酸萘酯(ANIT)所致肝损伤小鼠的保护作用。方法:将60只小鼠随机分为6组,分别为正常组、联苯双酯组、模型组、长毛风毛菊醇提物低、中、高剂量组(1.0,2.0和4.0 g·kg^(-1))。采用灌胃ANIT花生油溶液的方式复制肝损伤模型,造模48 h后小鼠眼静脉丛取血,测其血清中的丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、胆汁酸(TBA)、总胆红素(TBIL)、肿瘤坏死因子α(TNF-α)的含量;剖取3份肝脏分别用于检测肝组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的含量;分析对TNF-αmRNA表达的影响;观察肝组织病理形态学变化。结果:与模型组相比,长毛风毛菊醇提物中、高剂量组可显著降低肝损伤小鼠血清中ALT,AST,TBA,TBIL和TNF-α的含量(P<0.05,P<0.01);低剂量组只可显著降低肝损伤小鼠血清中TBIL,TNF-α的含量(P<0.05)。长毛风毛菊醇提物各剂量组均可使肝损伤小鼠肝匀浆中SOD水平显著升高,MDA水平显著下降(P<0.05,P<0.01);低、中剂量组能显著影响GSH-Px水平(P<0.01)。各给药剂量组均能影响肝损伤小鼠TNF-αmRNA的表达。病理切片结果显示长毛风毛菊醇提物各剂量组对肝组织的病理变化均有不同程度的改善作用。结论:长毛风毛菊醇提物对ANIT所致小鼠肝损伤有一定的保护作用。 Objective: To observe the protective effects of ethanolic extract of Saussurea. hieracioides Hook. F on mice liver injury induced by α-naphthyl isothiocyanate( ANIT). Methods: 60 mice were randomly divided into 6 groups,including normal group,bifendate group,model group,the low,medium,and high dose group of ethanolic extract of Saussurea. hieracioides Hook. F( 1. 0,2. 0,4. 0 g·kg^-1). The liver injury model was induced by lavage ANIT peanut oil solution. 48 hours after modeling,the serum was collected to detect the level of alanine aminotransferase( ALT),aspartate aminotransferase( AST),total bile acid( TBA),total bilirubin( TBIL)and tumor necrosis factor-α( TNF-α). The 3 liver samples were collected for the examination of the contents of malondialdehyde( MDA),glutathione peroxidase( GSH-Px) and superoxidase dismutase( SOD) in liver homogenate,the analysis of the impact of mRNA TNF-α expression,and the observation of hepaticpathological changes.Results: Compared with the model group,the medium and high dose group of ethanolic extract significantly decreased ALT,AST,TBA,TBIL,TNF-α in blood serum( P〈0. 05,P〈0. 01). The low dose group only hadevident effect on the TBIL,TNF-α in blood serum( P〈0. 05). All dose group remarkably advanced the activity of SOD and decreased the activity of MDA in liver homogenate( P〈0. 05,P〈0. 01). The low and medium dose group had significant effect on the GSH-Px level( P〈0. 01). The expression of mRNA TNF-α in mice with liver injury was affected by each dose group. All dose groups could ameliorate the hepatic pathological changes at different levels. Conclusion: the ethanolic extract of Saussurea. hieracioides Hook. F has a certain protective effect on mice liver injury induced by ANIT.
