摘要
探讨miR-581对结直肠癌SW620细胞侵袭转移能力的影响。首先,用miR-581mimics转染SW620细胞作为实验组(miR-581),用对照组mimics转染SW620细胞作为对照组(vector);用qRT-PCR检测2组细胞中miR-581的mRNA表达水平;划痕实验和transwell实验检测2组细胞迁移和侵袭能力的变化;Western blotting检测2组细胞中MMP-1和MMP-9的蛋白表达水平。结果显示,实验组细胞中miR-581表达较对照组明显增高(P<0.05);划痕实验显示实验组细胞划痕愈合率较对照组减慢(P<0.05),transwell结果显示实验组细胞较对照组细胞穿膜细胞数减少(P<0.05);Western blotting结果显示实验组MMP-1和MMP-9的表达都较对照组降低(P<0.05)。以上结果提示miR-581可以抑制SW620细胞的侵袭和转移。
We aimed to investigate the role of miR-581 overexpression in the invasion and metastasis of human colorectal cancer cell line SW620. SW620 cells were transfected with miR-581 mimics(miR-581) and the negative control mimics(vector). The mRNA expression of miR-581 was measured by qRT-PCR. Invasion and metastasis of the cells were detected by wound healingassay and transwell migration assays. The expression of MMP-1 and MMP-9 were detected by Western blotting. The results showed that the expression of miR-581 at mRNA level was significantly increased in the experimental group compared with that of the control group(P〈0.05). Up-regulation of miR-581 markedly enhanced SW620 cells" invasion and metastasis ability(P〈0.05). Western blotting showed that the expressions of MMP-1 and MMP-9 were significantly decreased in the experimental group(P〈0.05). These results suggest that overexpression of miR-581 inhibits the invasion and metastasis of colorectal cancer SW620 cells.
