摘要
目的通过对鞘氨醇激酶(Sph K1)基因敲除小鼠的繁殖和鉴定,以提供Sph K1缺失对各种疾病的影响的相关理想动物模型。方法将从美国国立健康研究院(National Institute of Health,NIH)处引进的Sph K1敲除杂合子小鼠进行交配,获得子代小鼠后,提取小鼠尾部的DNA,用琼脂糖电泳等方法检测及验证子代小鼠基因型。结果获得纯合子子代小鼠。结论 Sph K1基因敲除小鼠的鉴定获得成功。
Objective To provide an ideal animal model for the effect of SphK1 deletion on various diseases by the propagation and identification of sphingosine kinase(SphK1) knockout mice. Methods Heterozygote mice from NIH were bred and reproduced. Genome DNA was extracted from the murine tail and identified by agarose gel electrophoresis. Results SphK1 genotypes of the offspring mice included homozygous( SphK1-/-). Conclusion SphK1 knockout mice are successful reproduced.
