摘要
目的:探讨microRNA-138(miR-138)调控胶质瘤增殖和迁移能力的机制.方法:利用胶质瘤病例分析miR-138与胶质瘤临床病理之间的关系;利用realtime-PCR检测胶质瘤细胞U87和U251中miR-138的表达;MTT检测miR-138对胶质瘤细胞增殖的影响;划痕实验检测miR-138对胶质瘤细胞迁移的影响;Western blot检测miR-138对h TERT蛋白表达的影响.结果:MiR-138的表达水平与胶质瘤临床病理密切相关,胶质瘤患者普遍呈现miR-138低表达现象,miR-138在胶质瘤细胞U87和U251中的表达低于正常胶质细胞HEB.过表达miR-138抑制胶质瘤细胞U87和U251的增殖和迁移能力,同时miR-138能够下调h TERT蛋白的表达.结论:MiR-138通过下调h TERT蛋白的表达抑制胶质瘤细胞的增殖和迁移,是新的胶质瘤抑癌因子.
AIM : To study the mechanism of miRNA-138 ( miR- 138) regulating glioma proliferation and migration. METHODS: The relationship between miR-138 and glioma was analyzed. Real- time-PCR was uaed to detect the expression of miR-138 in U87 and U251 glioma cells ; MTT was used to detect the effect of miR- 138 on glioma cell proliferation; Would healing was used to detect the influence of miR-138 on glioma cell migration; Western blot was used to test the effect of miR-138 on hTERT protein expres- sion. RESULTS: The expression of miR-138 was closely related to the clinical pathology, and glioma patients exhibited miR-138 low expression phenomenon. MiR-138 was lower expression in U87 and U251 glioma ceils compared with normal glial cell HEB. MiR-138 inhibited U87 and U251 cell proliferation and migration. At the same time, miR-138 overexpression inhibited the expres- sion of hTERT. CONCLUSION: MiR-138 inhibits glioma cell proliferation and migration through down-regulating the expression of hTERT protein, it may he a new glioma tumor suppression factor.
