摘要
细胞凋亡是生物体内一种高度保守的死亡机制,而肿瘤的发生不仅与细胞增殖失控有关,也与细胞的凋亡减少有关,因此找到与肿瘤发生、发展密切相关的基因对靶向治疗肿瘤具有重要意义。生存素(survivin)是迄今为止发现的凋亡抑制蛋白家族中分子量最小、作用最强的凋亡抑制因子,由于其在恶性组织高表达,而在正常组织中表达几乎不表达,使其成为研究肿瘤标记和抗肿瘤治疗的标志蛋白。RNA干扰技术可以介导基因转录后水平的沉默,survivin-短发夹RNA(shRNA)可以作为一种新型的治疗肿瘤的方法应用于临床。
Apoptosis is a highly conserved death mechanism in vivo, and the occurrence of tumor is not only related to the uncontrolled proliferation of cells, but also related to the decrease of cell apoptosis. Therefore, it is of great significance to find the genes which are closely related to the occurrence and development of tumor. Survivin is so far found the smallest, the strongest apoptosis inhibitory factor of apoptosis inhibitory molecules protein family. Due to its high expression in malignant tissues, and almost no expression in normal tissues, it has become a tumor marker and anti-tumor therapeutic marker protein. RNA interference can be used to mediate gene silencing at the post transcriptional level, and survivin-shRNA can be used as a new method for the treatment of tumors in clinical.
