摘要
目的探讨二肽转运载体1(Pep T1)在大鼠结肠癌组织中的表达及其对营养免疫状况的影响。方法 Sprague-Dawley大鼠20只(术前组),随机分为研究组和对照组各10只,研究组通过移植皮下肿瘤瘤块建立大鼠结肠癌模型;对照组大鼠行腹部手术未种植肿瘤增生物,仅使盲肠暴露并提至腹外后还纳腹腔。检测三组大鼠结肠上皮Pep T1的表达情况、营养指标和T细胞免疫指标。结果与术前组相比,对照组大鼠肠黏膜组织仅Pep T1表达降低(P<0.05),而研究组肠黏膜组织Pep T1表达明显升高(P<0.05),总蛋白、白蛋白及前白蛋白水平、CD4+计数、CD8+计数、CD4+/CD8+比值均降低(P<0.05)。结论结肠癌大鼠出现营养不良及免疫功能异常,其机制可能与Pep T1在结肠癌组织中表达上调有关。
Objective To explore the expression of dipeptide transporter 1(Pep T1) in rat tissues with colon cancer and the influence of Pep T1 on the nutritional and immunological status of the rats. Methods Twenty Sprague-Dawley(SD) rats(preoperative group) were randomly divided into study group and control group,with 10 cases in each group. In the study group,rat model of colon cancer was developed by transplantation of subcutaneous tumor tissues. In the control group,abdominal surgery was performed,without tumor proliferation transplantation,which only made the cecum exposed and pulled to abdomen outside then was put back to the abdominal cavity. The expression of Pep T1,nutrition index and T lymphocyte immune index were detected in the three groups. Results Compared to the preoperative group,the expression of Pep T1 in cecum mucous tissues of rats decreased in the control group,but increased significantly in the study group(P〈0. 05),the levels of total protein,albumin and prealbumin,CD4+cell count,CD8+cell count and the ratio of CD4+to CD8+were lower in the study group(P〈0. 05).Conclusion Malnutrition and immunological abnormality occur in the rats with colon cancer,whose mechanism might be related to up-regulation of Pep T1 expression in colon cancer tissues.
