摘要
目的观察高、中、低三种剂量复健片治疗实验性脑梗死大鼠过程中对脑组织基质金属蛋白酶-2(MMP-2)、基质金属白酶-9(MMP-9)表达的变化,探讨复健片对星形细胞胶质化的影响。方法用Tamura法对右侧近端大脑中动脉进行电凝术以制备MCAO大鼠模型。按随机法将45只大鼠分为中药组、模型组以及假手术组,中药组又因复健片灌胃剂量的不同,分为中药高剂量组、中药中剂量组、中药低剂量组,其他两组予等量的蒸馏水。以上各组均为9只。3组MCAO大鼠分别在给药1周、2周、4周时处死后取脑组织,用酶联免疫吸附法测定大鼠脑组织MMP-2、MMP-9的表达。结果中药组治疗第2周和4周较1周比较,有统计学意义(P<0.05),与模型组同期比较有统计学意义(P<0.05),中药中剂量组较之同期高剂量组和低剂量组作用更为显著,尤其以第4周时最为明显。结论复健片改善实验性脑梗死大鼠神经修复的内环境,激活其内修复机制,通过上调MMP-2、MMP-9的表达而抑制星形细胞过度胶质化,以促进神经功能的重塑,是复健片治疗脑梗死的作用机制之一。
Objective To observe the effect of high,medium and low doses of Fujian tablets(FJT)on the expression of matrix metalloproteinase(MMP)-2 and MMP-9 in brain tissue in rats with experimental cerebral infarction,and explore its effect on astrocytes.Methods The middle cerebral artery occlusion(MCAO)rat model was prepared by Tamura method in the right proximal middle cerebral artery.Forty-five rats were divided into FJT group(high dose FJT group,middle dose FJT group and low dose FJT group),model group and sham operation group.Because of the different dosage of the Fujian tablet,the traditional Chinese medicine group was divided into the high dose group,middle dosegroup and low dose group,and the other two groups were given equal amount of distilled water.9 rats in each group.The MCAO rats in three groups were sacrificed at 1 week,2 weeks,4 weeks.The expressions of MMP-9 and MMP-2 in brain tissue were detected by ELISA mothed.Results After 2 weeks and 4 weeks,There were significant differences in the expressions of MMP-9 and MMP-2 in brain tissue between FJT groups and model group(P〈0.05).The expressions in middle dose FJT group were higher than that in high dose FJT group and low dose FJT group(P〈0.05),especially at fourth weeks.Conclusion FJT can up-regulated the expressions of MMP-2 and MMP-9,and inhibit the gliosis of astrocyte,promote the remodeling of neural function in in rats with experimental cerebral infarction.
