麝香对颅骨骨缺损模型大鼠骨组织转化生长因子β、血管内皮生长因子mRNA表达的影响 被引量:4

The Effect of Musk on mRNA Expression of Transforming Growth Factor β and Vascular Endothelial Growth Factor in Bone Tissues of Cranial Bone Defect Model Rats

摘要 目的探讨麝香促进骨缺损愈合的可能作用机制。方法 300只SD大鼠以颅骨钻制备颅骨骨缺损模型,造模后随机分为模型组和实验组,每组150只。两组再按照给药时间分为第7、14、28天3个时间点,每个时间点50只。实验组大鼠每天灌服天然麝香4.2 mg/100 g,模型组每天灌服等量的生理盐水,均每日灌胃1次。分别于灌胃第7、14、28天采用实时荧光定量PCR法检测两组大鼠骨缺损处组织中转化生长因子β(TGF-β)和血管内皮生长因子(VEGF)mRNA水平。结果与模型组同时间点比较,实验组第7、28天TGF-βmRNA及第28天VEGF mRNA表达显著升高(P<0.05);实验组第28天TGF-β、VEGF mRNA表达明显高于本组第14天(P<0.05)。结论麝香可有效促使大鼠颅骨骨缺损区的愈合,其机制可能与升高缺损处骨组织TGF-β、VEGF mRNA表达有关。 Objective To explore the possible mechanism of musk in improving bone defect healing. Methods Cranial bone defect models were made with cranial drill in 300 SD rats. After modeling,the rats were randomly divided into a model group and an experiment group,with 150 rats in each group. Rats in both groups were further divided into subgroups of 7 days,14 days,and 28 days,respectively,with 50 rats in each subgroup. The experiment group was gavaged with 4. 2 mg/100 g of natural musk,while the model group was gavaged with equivalent saline,once per day for 7 days,14 days or 28 days,respectively. On the 7th,14 th and 28 th day of gavage,real-time fluorescent quantitative PCR was adopted to detect levels of transforming growth factor β( TGF-β) mRNA and vascular endothelial growth factor( VEGF) mRNA in bone defect regions in both groups. Results Compared with those in the model group at the same time point,level of TGF-β mRNA on the 7th and the 28 th day and level of VEGF mRNA on the28 th day in the experiment group increased remarkably( P〈 0. 05). In the experiment group,expression of TGF-βmRNA and VEGF mRNA on the 28 th day were remarkably higher than that on the 14 th day( P〈 0. 05). Conclusion Musk could effectively improve healing of cranial bone defect regions. Its mechanism may be related to increasing of expression of TGF-β mRNA and VEGF mRNA in bone tissue in defect regions.
