摘要
目的:探讨提高去白细胞悬浮红细胞储存期末血液质量的最佳离心条件。方法:选取2016年采集的全血1 500袋(每袋400 ml),随机分成A、B、C组制成去白细胞悬浮红细胞。离心条件分别为A组4℃,3 000 r/min,离心7 min;B组4℃,3 600 r/min,离心5 min;C组4℃,3 600 r/min,离心12 min。对3组去白细胞悬浮红细胞储存期末血液质量进行检测,并对结果进行比较。结果:3组储存期末溶血率达标率分别为97.4%、96.4%、92.2%,A组和B组比较差异无统计学意义,A组和B组与C组比较差异有统计学意义(P<0.05)。血细胞比容达标率,A组和C组比较差异无统计学意义,A组和C组与B组比较差异有统计学意义(P<0.05)。结论:去白细胞悬浮红细胞在制备过程中离心条件与红细胞溶血程度有一定关系,为保证去白细胞悬浮红细胞的质量,降低去白细胞悬浮红细胞储存期末溶血率,血液滤除白细胞后建议选择低速离心。
Objective: To investigate the best condition of centrifugation on increasing the blood quality of terminal storage of leukocyte-removed red cells suspension. Methods: 1 500 sack plasma(400 ml a sack)which collected in 2016,assigned to A B C three groups randomly to make leukocyte-removed red cells suspension. Centrifugation conditions was as follows, group A 4 ℃ 、 3 000 r/min、 7 minutes centrifugation,group B 4 ℃ 、3 600 r/min、5 minutes centrifugation,group C 4 ℃ 、3 600 r/min、12minutes centrifugation. The terminal storage blood quality of the three group of leukocyte-removed red cells suspension were detected and the results were compared. Results: The hemolysis standard-reaching rate of the three groups terminal storage were 97.4%、96.4%、92.2%,the comparative difference of group A and group B had no statistics means,compared A with B and C had statistics means(P〈0. 05). Blood cells specific volume standard-reaching rate,the comparative difference of group A and group C had no statistics means,compared A with C and B had statistics means(P〈0. 05). Conclusion: In the process of making leukocyte-removed red cells suspension,centrifugation conditions have connected with hemolysis degree,in order to guarantee the quality of leukocyte-removed red cells suspension and reduce the hemolysis rate of terminal storage leukocyte-removed red cells suspension,the low speed centrifugation are suggested choice when filtering leukocyte in blood.
