利用转录组测序技术分析ADC和NOS基因mRNA在鹅等级前卵泡中的表达 被引量:1

mRNA Expression of ADC and NOS Genes mRNA in Pre-hierarchical Follicles of Geese by Transcriptome Analysis

摘要 为研究精氨酸代谢的关键酶精氨酸脱羧酶(Arginine Decarboxylase,ADC)和一氧化氮合酶(Nitric-oxide Synthase,NOS)mRNA在鹅等级前卵泡中的表达情况。采用高通量测序技术对鹅等级前卵泡进行转录组测序分析,并通过荧光定量PCR对结果进行验证。通过FPKM值计算分析,这2个基因在鹅等级前卵泡中均有表达,且随着卵泡的发育,NOS mRNA的表达呈现先降低后增加再降低的趋势,在初级卵泡中的表达量达到最高值,在中白卵泡中的表达量最低。ADC mRNA的表达则呈现先增加后降低再增加的趋势,在初级卵泡中的表达量最低,在小白卵泡中的表达量最高。荧光定量PCR结果与转录组测序结果基本相同。 We used transcriptome sequencing technology to study the expression of key enzymes of polyamine metabolism,arginine decarboxylase( ADC) and nitric-oxide synthase( NOS) in pre-hierarchical follicles of geese. Transcriptome of the geese pre-hierarchical follicles was analyzed by using Illumina Hiseq 2500 and differential expression of ADC and NOS mRNA in the sampled follicles was confirmed by fluorescence quantitative PCR. The results showed that through computational analysis of FPKM value the genes were all expressed in pre-hierarchical follicles. With the development of follicles,mRNA expression of NOS gene displayed firstly a decrease trend and then an increase-decrease trend. At the primary follicles,NOS had the highest expression. At the middle white follicles,its expression was the lowest. The expression mRNA of ADC displayed firstly an increase trend and then a decrease-increase trend. At the primary follicles,NOS had the minimum expression. At thesmall white follicles,its expression was the highest. The transcriptome results are basically consistent with those of fluorescent quantitative PCR.
