摘要
目的探讨miR-155及COX-2在肝细胞癌(HCC)组织中的表达及其与肝癌患者术后2年复发率的关系。方法采用免疫组化法检测67例肝细胞癌组织及癌旁肝组织中COX-2蛋白表达情况;实时荧光定量RT-PCR方法检测miR-155的表达,分析两者与肝细胞癌患者术后2年复发率的关系。结果免疫组化结果显示67例肝细胞癌组织中有41例(61.2%)COX-2蛋白表达阳性,实时荧光定量RTPCR结果显示44例(65.6%)肝细胞癌组织中miR-155表达升高,高于癌旁肝组织(P<0.001)。miR-155高表达同时COX-2表达阳性的患者术后2年复发率明显高于miR-155低表达同时COX-2阴性的患者(86.2%vs 34.8%,P=0.001)。多因素分析(Cox模型)显示:Edmonson病理分级(P=0.027)、TNM分期(P=0.011)以及miR-155联合COX-2(P=0.026)是影响HCC患者术后2年复发率的独立危险因素。结论miR-155与COX-2蛋白在HCC组织中表达上调,两个指标联合检测对HCC术后预后有预测作用。
Objective To investigate the expression of miR-155 and COX-2 in hepatocellular carcinoma(HCC),and the relationship between the two factors and the 2-year postoperative recurrence rate of HCC.Methods The expression of COX-2 protein in HCC tissues and the adjacent non-tumor tissues of 67 cases was detected by immunohistochemistry; the expression of miR-155 was detected by real-time fluorescent quantitative RT-PCR. Then analyzed the relationship between the two factors and their relationship with the 2-year postoperative recurrence rate of HCC.Results The immunohistochemical stainings showed that 41 of 67(61.2%)cases were positive expression in HCC tissues. And the results of real-time quantitative RT-PCR showed that the expression of miR-155 in HCC were higher than the paracancerous liver tissue in 44(65.6%) cases(P0.001).The patients with higher expression of miR-155 and positive expression of COX-2 protein had a higher 2-year postoperative recurrence rate than the patients with lower expression of miR-155 and negative expression of COX-2 protein(86.2% vs 34.8%,P = 0.001). Multivariate analysis(Cox model) showed that Edmonson pathological grade(P = 0.027),TNM stage(P = 0.011) and miR-155 combined with COX-2(P = 0.026) were independent risk factors for the 2-year postoperative recurrence rate of HCC patients.Conclusions The expression of miR-155 and COX-2 protein is up-regulated in HCC tissues,combined detection of the two indexes can be used for evaluating the prognosis of postoperative patients with HCC.
