摘要
目的探究耐奥沙利铂人胃癌SGC-7901细胞株生物学特性与上皮问质转化(epithelial-mesenchymal transition,EMT)之间的关系。方法采用体外奥沙利铂低浓度逐渐递增法间断作用人胃癌SGC-7901细胞株,建立耐药细胞株;通过形态学、MTT法、Transwell实验分别检测SGC-7901细胞株和耐药细胞株的形态特征、增殖能力、药物敏感性和侵袭转移能力;通过免疫荧光染色检测波形蛋白、上皮钙粘附分子在两组细胞株之间的表达差异。结果与原代SGC-7901细胞株相比,耐奥沙利铂细胞株呈间质样细胞表型,增殖能力减弱,对奥沙利铂敏感性明显降低;转移侵袭能力增强;波形蛋白水平上调,上皮钙粘附分子水平下降。结论耐奥沙利铂人胃癌SGC-7901细胞株增殖能力减弱,但具有高侵袭性及EMT特征。
Objective To study the biological characteristics of the oxaliplatin-resistant human gastric cancer cell line SGC-7901 and their relationship with epithelial-mesenchymal transition (EMT). Method Human gastric cancer SGC-7901 cells were intermittently exposed to L-OHP in manner from low to high concentration in vitro to establish the L-OHP-resistant cell line. The morphological charac-teristics, proliferation ability, drug sensitivity, invasion and metastasis of the SGC-7901 group and the drug-resistant group were detected by morphological examination, MTT assay and transwell experiment respectively. The expression of vimentin and E-cadherin in this two groups of cell lines was detected and compared by immunofluorescence. Results Compared to that of the primary SGC-7901 cell line, the morphology of the L-OHP-resistant cell line was more like mesenchymal cells with decreased proliferation ability, the sensitivity to ox- aliplatin significantly reduced, but the abilities of invasion and metastasis enhanced, vimentin expression up-regulated, and E-cadherin expression declined. Conclusion The proliferation ability was decreased in oxaliplatin-resistant human gastric cancer cell line SGC- 7901 compared to that in the non-reisistant one, but the abilities of invasion and metastasis were enhanced with EMT characteristics.
