非小细胞肺癌组织中COX-2和血管内皮生长因子免疫反应性增强而E-钙粘素免疫反应性减弱 被引量:3

摘要 目的检测非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中环氧化酶-2(cyclooxygenase-2,COX-2)、血管内皮生长因子(vascular endothelial growth factor,VEGF)和E-钙粘素(E-cadherin,E-Cad)的表达,并分析它们之间的相互关系以及临床病理意义。方法应用免疫组织化学法检测63例NSCL£组织和20例肺良性疾病对照组中COX-2、VEGF、E-cad的免疫反应性。结果 COX-2和VEGF在NSCLC组织中的免疫反应阳性率显著高于对照组,E-cad在NSCLC组织中的免疫反应阳性率显著低于对照组。NSCLC中,COX-2和VEGF的免疫反应阳性率与淋巴结转移之间呈正相关,而且COX-2在肺腺癌组织中的免疫反应阳性率显著高于肺鳞癌。E-cad的免疫反应阳性率与NSCLC组织的分化程度、淋巴结转移及临床分期之间呈负相关。Spearman相关分析显示,NSCLC组织中E-cad与COX-2和VEGF的免疫反应性之间呈负相关,COX-2与VEGF的免疫反应性呈正相关。结论 COX-2、VEGF的高表达及E-cad的低表达可能在NSCLC的发生、浸润、转移中发挥重要作用,联合检测其表达对判断NSCLC的临床进展、推测预后及制定靶向治疗方案有一定的参考价值。 Objective To investigate the expression of COX-2, VEGF and E- cad as well as their correlation and clinic patho-logical significance in non-small cell lung cancer (NSCLC). Methods The immunoreactivity of COX-2, VEGF and E-cad in 63 cases of NSCLC and 20 cases of benign lung lesion control tissue was detected by immunohistochemical staining. Results The positive immu- noreactive rates of COX-2 and YEGF were significantly higher, and that of E-cad significantly lower in the NSCLC group than those in the control group. In NSCLC, the immunoreactive rates of COX-2 and VEGF were positively correlated with lymphatic metastasis; mo-reover, the immunoreactive rate of COX-2 was significantly higher in pulmonary adenocarcinoma tissues than in pulmonary squamous cell carcinoma. The immunoreactivity of E-cad was negatively correlated with the differentiation grade of NSCLC tissue, lymphatic node metastasis, as well as TNM stages. Spearman analysis showed a negative correlation between the immunoreactivity of E-cad and that of COX-2 or YEGF, and a positive correlation between the immunoreactivity of COX-2 and that of VEGF. Conclusion The high expres-sion of GOX-2 and VEGF and low expression of E-cad may play an important role in the pathogenesis, invasion and metastasis of NSCLC. To comprehensively test their expression can provide information for judging the clinical progress of NSCLC, calculating prog-nosis and making out targeted therapy protocols.
