摘要 定向进化技术是近二十年发展起来的蛋白质改造技术,其可通过对基因的突变和重组来构建突变库,然后通过高通量的筛选鉴定性能改善的突变体,此技术是改善酶性能最有效的途径之一。本文就定向进化技术的几种常用方法和定向进化后的文库筛选、应用及展望进行了概述。 Directed evolution method has been a kind of new technology of protein modification developed in recent 20 years,which build mutant library through the gene mutation and restructuring,and then identified the performance improvement of mutants with high-throughput screening methods.This was the effective way to improve enzyme properties.This paper reviewed the several frequently-used methods of directed evolution and library screening after directed evolution of protein.In addition,the application of recombination for directed evolution and prospection for the future of this technology also described.
