白三叶新品系无菌苗培养及愈伤组织诱导

摘要 采用多因素区组试验方法,以白三叶新品系种子为材料对白三叶新品系无菌苗培养过程中消毒方法、培养基配方进行研究,以培育的无菌苗为材料对不同植物生长调节剂和不同外植体对白三叶新品系愈伤组织诱导的影响进行研究。结果表明:采用70%酒精消毒3min+2%NaClO消毒10min,种子消毒效果最佳且对种子毒害作用最小,且在添加1.5%蔗糖的MS培养基上最适合无菌苗培养;白三叶新品系的茎较易脱分化,以茎为外植体进行愈伤组织培养,最佳培养基为MS+2ug·L^(-1)6-BA+3ug·L^(-1)2,4D,以叶为外植体进行愈伤组织培养,最佳培养基为MS+0.5ug·L^(-1)6-BA+1ug·L^(-1)2,4D。 With rnultifactor block test method, disinfection methods for seed, medium formula for sterile seed- lings and callus of new hybrid Trifoliurn repens L. were researched, the results showed that using 70% alcohol for 3 minutes and 2~/00 NaC10 for 10 minutes were the best seed disinfection method and had the least toxic to the seed. The most suitable medium for sterile seedling culture was MS supplemented with 1.5% sucrose. The stem of new hybrid Trifolium repens L. was more easier to be dedifferentiated, and the best callus induction medium for stem was MS+2 ug · L ^-1 6-BA+3 ug · L^-1 2,4D, the best callus induction medium for leaf was MS+0.5 ug· L^-1 6-BA+1ug· L^-1 2,4D.
