摘要 目的研究MUC5AC(黏蛋白MUC5AC)和TNF-α(肿瘤坏死因子-α)在大鼠中耳炎黏膜的表达变化。方法40只SD大鼠随机分为对照组10只、实验组30只;实验组建立大鼠急性分泌性中耳炎模型,采用q PCR检测MUC5AC及TNF-α在第1天,第3天,第7天两组中耳黏膜的表达变化,并用免疫组织化学染色技术分别检测MUC5AC和TNF-α在第1天,第3天,第7天在中耳黏膜的表达变化。结果在对照组中,MUC5AC不表达,TNF-α轻度表达,实验组从第1天到第3天MUC5AC及TNF-α的表达随着时间的延长而增强,至第7天则表达开始减弱,实验组与对照组的比较差别具有统计学意义(P<0.05)。结论 MUC5AC和TNF-α在大鼠急性分泌性中耳炎黏膜中表达上调,且与时间相关。 Objective To study expression of MUC5 AC and TNF-α in the middle ear mucosa in rat model of acute otitis media with effusion.Methods Forty adult female Sprague-Dawlay(SD) rats were randomly divided into a control group and a experimental group.Rats in the experimental group received injection of endotoxin.The level and pattern of MUC5 AC and TNF-α expression within the middle ear epithelium were tested by q PCR and immunohistochemical assays.Results q PCR and immunohistochemical staining results showed increased expression of MUC5 AC and TNF-α in the experimental group along with time(*P〈0.05) and the MUC5 AC were mainly concentrated on the goblet cells.Conclusions Expression of MUC5 AC and TNF-α is increased in rat middle ear epithelium with acute media otitis with effusion.Hence,it is speculated that MUC5 AC and TNF-α may participate in the process of acute media otitis with effusion.
