副干酪乳杆菌VL8胞外多糖协同ConA对小鼠脾淋巴细胞增殖及细胞因子分泌的影响 被引量:1

摘要 本研究以前期从viili中分离得到的一株副干酪乳杆菌VL8为基础,对其胞外多糖(VL8-EPS)的免疫活性进行了初步探究。首先分离制备小鼠脾淋巴细胞,采用四甲基偶氮唑盐(MTT)法检测不同浓度的VL8-EPS对小鼠脾淋巴细胞增殖及在其与有丝分裂原共同作用下对小鼠脾淋巴细胞的增殖作用;双抗体夹心酶联免疫吸附实验(ELISA)法检测细胞因子白介素-2(IL-2)、干扰素-γ(IFN-γ)、白介素-6(IL-6)的含量。结果表明,VL8-EPS在浓度12.5~800μg/m L范围内,能显著促进小鼠脾淋巴细胞增殖;VL8-EPS协同刀豆蛋白(Con A)也显著地增强了脾淋巴细胞的增殖作用,且诱导效果高于单独施用VL8-EPS;并能显著促进协同Con A诱导的细胞因子IL-2、IFN-γ、IL-6的分泌水平。综上,VL8-EPS显著地促进了脾淋巴细胞的增殖,并对Con A诱导的脾淋巴细胞的增殖有协同促进作用,还通过促进细胞因子的分泌发挥了免疫增强作用,说明VL8-EPS在一定程度上对免疫机制进行了调控。 The strains of lactobacillus paracasei VL8 was isolated form viili as base as described previously,luther researching of the immunomodulation of VL8-EPS was investigated.Firstly,preparation of mouse spleen lymphocytes and the proliferation of spleen lymphocytes in mice with different concentrations of VLS-EPS were determined by MTI'.The contents of IL-2, IFN-γ and IL-6 were determined by using ELISA.The results showed that VL8-EPS at a concentration ranging from 12.5 μg/mL to 800 μg/mL resulted in a significant increase of spleen lymphocyte proliferation, VL8- EPS could also enhance the effect of ConA-induced spleen lymphocyte proliferation and the induction effects was higher than that of single application of VL8- EPS.Moreover, VLS- EPS could significantly increase the secretion level of IL- 2, IFN- y and IL- 6 in ConA- induced spleen lymphocyte. In summary,VL8-EPS can effectively increase spleen lymphocyte proliferation and enhance the effect of ConA- induced spleen cell growth. VL8-EPS has the ability to enhance immune responses by increasing the secretion of cytokine.lt showed that VL8-EPS could regulate the immune mechanisms partly.
