淫羊藿苷对3T3-L1前脂肪细胞增殖及分化的影响 被引量:4

摘要 目的研究淫羊藿苷(ICA)不同作用浓度及不同作用时间对3T3-L1前脂肪细胞增殖和分化的抑制作用。方法采用MTT法观察ICA对小鼠3T3-L1前脂肪细胞增殖的影响;应用ICA诱导小鼠3T3-L1前脂肪细胞分化,油红O染色测定细胞内脂质含量,观察ICA对小鼠3T3-L1前脂肪细胞分化的影响。结果 67μg/m L ICA对细胞增殖起抑制作用,持续至96 h并达到最大;30μg/m L和15μg/m L ICA在72 h、96 h对细胞增殖起抑制作用。结论较高浓度ICA对3T3-L1前脂肪细胞的分化有抑制作用,而低浓度ICA对3T3-L1前脂肪细胞的分化作用不明显。 Objective It is to explore the inhibitory effect of different concentration and action time of icariin on the proliferation and differentiation of 3T3-L1 preadipocytes and its possible mechanism. Methods MTT assay was used to explore the effect of icariin on the proliferation of mouse 3T3-L1 preadipocytes. Differentiation of mouse 3T3-L1 preadipocytes was induced by icariin. Oil red O staining quantitative method was used to determination the fat content in the cells,in order to observe the effect of icariin on the differentiation of mouse 3T3-L1 preadipocytes. Results ICA inhibited cell proliferation at a concentration of 67 μg/m L,the inhibitory effect lasted for 96 h,then reached its maximum; ICA at the concentration of 30,15 μg/m L,could inhibit the differentiation of preadipocytes when the action time lasted from 72 h to 96 h. Conclusion ICA can inhibit the proliferation of 3T3-L1 preadipocytes,and there is a dose-dependent. ICA higher concentrations can inhibit 3T3-L1 preadipocytes differentiated,however,the effect of low concentrations ICA was not significant.
