组织蛋白酶K与肿瘤生长转移相关性的研究进展

摘要 组织蛋白酶K,又称溶酶体半胱氨酸组织蛋白酶K,是木瓜半胱氨酸蛋白酶家族的一员,含329个氨基酸。主要底物是Ⅰ型胶原,可参与细胞外基质重建,也能降解骨骼基质中的其他蛋白酶,如骨连接蛋白、血管内皮生长因子,共同促进肿瘤转移。本文综述了组织蛋白酶K在前列腺癌、乳腺癌、黑色素瘤、鳞状细胞癌等多种肿瘤生长、侵袭和转移中的影响与作用,揭示了Cat K与多种肿瘤疾病的相关性,对恶性肿瘤的诊断和预后治疗具有一定的临床意义。 Cathepsin K belongs to the papain super family of lysosomal cystein protease,containing 329 amino acids. Cathepsin K can effectively degrade native collagen I,involved in the reconstruction of extracellular matrix and also degrade other proteases in the bone matrix,such as SPARC,VEGF,to promote tumor metastasis.This paper reviews the effects of cathepsin K on the growth,invasion and metastasis of prostate cancer,breast cancer,melanoma,squamous cell carcinoma and other tumors. It reveals the correlation between Cat K and tumor diseases,and may have a clinical significance in tumor diagnosis and prognosis.
