摘要
目的研究小檗碱在小鼠脑缺血恢复期的作用,及其对胶质细胞的影响。方法建立小鼠脑缺血模型,造模当日开始腹腔注射小檗碱并持续给药14天。通过转棒实验评价动物的神经功能,通过脑指数评价脑萎缩程度。利用体外培养的胶质细胞,给予10μM小檗碱处理24h,观察对细胞增殖、细胞周期和凋亡的影响。结果小檗碱能减轻脑萎缩,促进小鼠脑缺血损伤后神经功能的恢复。体外实验证明小檗碱抑制胶质细胞的增殖,诱导了细胞周期阻滞和凋亡。结论小檗碱促进小鼠脑缺血损伤后的修复,可能是通过对胶质细胞的影响发挥神经保护作用。
Objective To investigate the role of berberine in the recovery of cerebral ischemia in mice and the neuroprolection mechanism mediated by glial cells. Methods The stroke mice were induced via middle cerebral artery occhlsion and intraperitoneal injected of berberine daily until euthanized on d14. The neurological function was assessed by rotarod test and the brain atrophy was assessed by cerebral index. Glial cells cuhured in vitro were stimulated by berberine with the concertratiou of 10μ M for 24h. Then cell viability was analyzed by CCK-8 kit. Furthermore, cell cycle and apoptosis were analyzed by flow cytometry. Results Berberine was confirmed to reduce brain atrophy in mice and promote the recovery of neurological function after cerebral ischemia injury. In addition, in vitro studies showed that berberine suppressed the proliferation of BV2 microglial cells, as well as arrested U87 aslroglioma cells at GO/GI phase and induced cell apoplosis. Conclusion Berberine exerts nenroprotection in the recoverv of cerebral ischemia, possibly as a resuh of the suppression of proliferation and induction of apoptosis in glial cells.
