新肥菇原生地土壤有机碳氮测定条件优化

摘要 为优化新肥菇原生地土壤有机碳氮的测定条件,获取准确的碳氮含量,为该野生食用菌的栽培料营养配比提供科学依据,以采自新疆的新肥菇原生地土壤为试验材料,分别研究了浸提剂、浸提液浓度、样液比和浸提时间对土壤可溶性有机碳氮测定的影响。结果表明:浸提剂和浸提剂浓度对土壤可溶性有机碳的测定有重要影响,样液比和浸提时间对测定影响不显著。而浸提剂、浸提液浓度、样液比和浸提时间对土壤可溶性总氮的测定均无显著影响。选用新鲜土壤,在室温下以1.5 mol·L-1K2SO4为浸提剂,样液比1∶4,浸提时间20 min时测定结果重复性好,精确度高,符合检测要求。 In order to optimize the determination conditions of soil organic carbon and nitrogen in Agaricus sinodeliciosus habitat and provide supports on the cultivation material ratio of nutrition, dissolved total organic carbon and total nitrogen of soil collected in the habitat were measured with different extracting agents, concentrations, water/soil ratios, and extraction time.Results showed that extracting agent and concentration had an important effect on the determination of soil dissolved organic carbon, but the water/soil ratio and extraction time showed no significant influence on the measured value.However, extracting agents, concentrations, water/siol ratios, and extraction time had no significant influence on the determination of soil dissolved total nitrogen.The suitable protocol was using fresh soil as the determination subject under condition of room temperature, 1.5 mol·L-1 K2SO4, 1∶4 water/soil ratio, and extraction for 20 min.
