摘要
目的探讨夏枯草水提物的抗抑郁作用及可能机制。方法小鼠分为正常组,模型组,盐酸氟西汀(阳性对照药,0.02 g·kg^(-1))组,夏枯草水提物低、中、高剂量(2,4,8 g·kg^(-1))组,给药组连续8 d灌胃给药。末次给药1 h后,采用小鼠悬尾、强迫游泳以及自主活动试验对小鼠进行行为学观察,并采用ELISA法测定小鼠海马组织5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)、脑源性神经营养因子(BDNF)、环氧合酶-2(COX-2)、前列腺素E_2(PGE_2)、白细胞介素-1β(IL^(-1)β)和白细胞介素-6(IL-6)的含量。结果各给药组与正常组比较,对小鼠自主活动无明显影响,差异无统计学意义(P>0.05);与模型组比较,夏枯草水提物中、高剂量组能显著缩短小鼠悬尾和强迫游泳不动时间(P<0.001),增加小鼠海马组织中5-HT、NE、DA的含量(P<0.05,P<0.01),降低小鼠海马组织中COX-2、PGE_2、IL^(-1)β、IL-6的含量(P<0.01,P<0.001)。结论夏枯草水提物可能通过提高海马组织中单胺类神经递质的含量、降低炎症因子的含量来产生抗抑郁作用。
Objective To study the antidepressant effects of the water extracts from Prunella vulgaris L. in mice and elucidate the mechanism of antidepressant. Methods The mice were divided into normal control group, model control group, and four treatment groups. The treatment groups received fluoxetine (positive control) at a dose of 0.02 g·kg^-1 and water extracts of Prunella vulgaris L. at doses of 2, 4, 8g·kg^-1 for 8 days, respectively. Tail suspension lest (TST), forced swimming test (FST), and spontaneous motion test were conducted for behavior evaluation 1 h after the last administration. The contents of serotonin (5-HT), norepinephrine (NE), dopamine (DA) and brain derived neurotrophic factor (BDNF), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), Interleukin- 1 beta (IL- 1β) and Interleukin-6 (IL-6) in the hippocampus of mice were determined by ELISA. Results Compared with the normal group, treatment groups had no effect on spontaneous motion (P 〉 0.05 ). Compared with the model group, middle- and high-dose groups of the water extracts of Prunella vulgaris L. significantly decreased immobility time of mice in both TST and FST(P 〈 0.001) , increased the contents of 5-HT, NE, DA(P 〈 0.05, P 〈 0.01) , and decreased the contents of COX-2, PGE2, IL-1β, IL-6 in the hippocampus of mice (P 〈 0.01, P 〈 0.001). Conclusion These results suggested that water extracts of Prunella vulgaris L. possesses potent antidepressant-like effect by increasing the content of monoamine neurotransmitters and decreasing the content of inflammatory factors.
