摘要
建立了电化学循环伏安法测定葡萄糖氧化酶活性的方法。该方法引入二茂铁甲醇作为氧化还原探针,以玻碳电极作为工作电极、饱和甘汞电极作为参比电极、铂丝电极作为辅助电极,采用循环伏安法,得到不同酶活的峰电流值,在酶浓度为0.128~2.377U/mL,酶浓度与峰电流之间存在线性关系,相关系数为0.998,检测下限为0.128 U/mL。运用该法测定了孢子萌发过程中酶活的变化,随着孢子的萌发,酶活先增加,达到最大值(2.130U/mL)后再次下降,同时采用传统滴定法进行对照。结果表明,相较于传统方法,电化学方法具有准确、快速和简便的特点。
In this study, the electrochemical cyclic vohammetry method was developed for the determination of activity of glucose ox- idase. The ferroeene methanol was introducing as the redox probes on cyclic voltammetry, using glassy carbon electrode as working elec- trode, saturated calomel electrode as reference electrode, platinum wire electrode as the counter electrode and the peak current value of different enzyme activity was required . There was a linear relation- ship between the peak current in enzyme concentration of 1.282 2.377 U/mL with the correlation coefficient as 0.998, the detection limit as 1.282 U/mL. The enzyme activity of spores from dormancy to germination was detected by this new method, as the enzyme ac- tivity of spores decreased after increasing first, reaching the maximum (2.130 U/mL). The traditional methods were tested at the same time. Compared with the traditional method, the proposed elec- trochemical method has the characteristics of accurate, fast and con- venient.
