布渣叶提取物促高脂血症家兔胆固醇逆向转运机制研究 被引量:4

The Mechanism of Microcos Paniculata Extract on RCT in Hyperlipidemia Rabbits

摘要 目的:研究布渣叶提取物(MP)的调血脂作用及胆固醇逆向转运(RCT)机理。方法:复制食饵性高脂血症家兔,给予MP(1.4、2.8和5.8g/kg)连续给药6周,检测血清、肝脂质TC、TG、HDL-C和LDL-C含量,肝ApoA1、CYP7A1、LXR-α、LPL及回肠NPCL1L1 mRNA和蛋白表达。结果:MP可显著降低高脂血症家兔血清和肝脏TC、TG(P<0.05),增强肝、回肠ApoA1、CYP7A1、LXR-αand LPL mRNA和蛋白表达(P<0.05),抑制NPC1L1 mRNA和蛋白表达(P<0.05)。结论:MP可通过促进RCT、减少内外源性胆固醇及代谢产物吸收或再吸收,从而调血脂、逆转肝脂肪变性。 Objective: To investigate the lipid regulating effect of Microcos paniculata (MP) extract and the mechanism of reverse cholesterol trans- port (RCT). Methods: Copied diet-induced hyperlipidemia rabbits and continuously administrated with MP (1.4, 2.8 and 5.8g/kg) for 6 weeks, then detected their contents of serum and hepatic lipid TC, TG, HDL-C and LDL-C, and the mRNA and protein expressions ofApoA1, CYP7A1, LXR-ct, LPL in liver and NPCL1L1 in ileum. Results: MP could significantly reduce the serum and liver TC and TG of hyperlipidemia rabbits (P〈0.05), en- hance their mRNA and protein expressions of ApoA1, CYP7A1, LXR-ct and LPL (P〈0.05), inhibit the expressions of NPC1L1 mRNA and protein (P〈0.05). Conclusion: MP can regulate lipid and reverse liver fatty degeneration via promoting RCT and reducing the absorption or resorption of en- dogenous and exogenous cholesterol and their metabolites.
