益甘宁颗粒对对乙酰氨基酚诱导小鼠肝损伤模型的保护作用 被引量:5

摘要 目的探讨益甘宁颗粒对对乙酰氨基酚(acetaminophen,APAP)诱导的小鼠肝损伤的保护作用以及作用机制。方法 60只小鼠随机分为6组,分为正常组、模型组、联苯双酯组(阳性对照)、益甘宁颗粒低剂量组(0.13 g·m L^(-1))、益甘宁颗粒中剂量组(0.39 g·m L^(-1))和益甘宁颗粒高剂量组(1.17g·m L^(-1)),各剂量组小鼠连续灌胃益甘宁颗粒14 d。以一次性腹腔注射APAP 300 mg·kg^(-1)诱导肝损伤模型,ELISA法测定小鼠血清中丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)的浓度,病理切片观察肝组织的损伤情况,硫代巴比妥酸检测丙二醛(MDA)含量;黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)活性。结果与模型组相比,益甘宁颗粒各剂量组小鼠血清中AST、ALT的浓度均降低;肝脏较完整、清晰,只有少量细胞坏死;MDA含量显著降低(P<0.01);SOD活性较模型组显著升高(P<0.01)。结论益甘宁颗粒对APAP损伤的肝细胞具有很好的保护作用,这可能与其抗自由基有关。 Objective To determine the protective effect of Yiganning granules on the murine liver jnjury induced by acetaminophen(APAP). Methods Sixty mice were randomly divided into a control group, a model group, a bifendate group(positive control group), and 3 Yiganning granules groups at 0.13, 0.39, and 1.17 g·mL^-1. After 14 d, all the mice were intraperitoneally given APAP at 300 mg·kg^-1 to induce liver injury. The concentrations of ALT and AST were assessed by ELISA, liver damage was determined, the MDA concentration was observed by TBA method, and the level of SOD was tested by xanthine oxidase. Results Compared with the model group, each Yiganning granule group had reduced AST and ALT levels. In the liver of the mice treated with different doses of Yiganning granules, the lobule structure of the liver was intact and clear, and necrosis was seen in only a few hepatocytes. The concentration of MDA was reduced and SOD activity was significantly increased compared with those of the model group. Conclusion Yiganning granules have protective effect on murine liver injury were induced by APAP via probablly eliminating free radicals in mice.
