藤珠胃康颗粒中4种皂苷成分的大鼠体内药代动力学研究 被引量:4

摘要 目的研究藤珠胃康颗粒中人参皂苷Re、Ro、Rb1和竹节参皂苷Ⅳa在大鼠体内的药代动力学特征。方法大鼠灌胃藤珠胃康颗粒(9 g·kg^(-1)),于不同时间点眼眶静脉丛采血,乙腈沉淀蛋白,采用HPLC-MS法检测大鼠血浆中的人参皂苷Re、Ro、Rb1和竹节参皂苷Ⅳa,应用DAS 2.1.1软件拟合药动学参数。结果人参皂苷Re、Ro、Rb1和竹节参皂苷Ⅳa的t_(1/2)分别为(13.72±9.14)、(6.94±3.51)、(28.80±9.99)、(5.17±2.41)h,tmax分别为(1±0.00)、(4±0.00)、(2±0.00)、(4±0.00)h,Cmax分别为(370.27±88.27)、(649.06±98.22)、(111.10±3.38)、(686.94±145.55)ng·m L^(-1),AUC0~t分别为(2188.75±599.81)、(4743.44±509.26)、(1699.06±35.66)、(4427.82±1396.29)μg·h·L^(-1)。结论建立的人参皂苷Re、Ro、Rb1和竹节参皂苷Ⅳa血浆样品分析方法及得到的药动学参数,可为藤珠胃康颗粒的药代动力学研究提供参考。 Objective To determine the pharmacokinetics of 4 saponins (ginsenoside Re, ginsenoside Ro, gin- senoside Rbl and chikusetsu saponin lVa) in Tengzhu Weikang granules in rats. Methods After different time points of Ig administration of 9 g kg-1 Tengzhu Weikang granules, blood was taken from the venous plexus of 0.indus and the plasma was precipitated with acetonitrile. HPLC-MS was used to measure ginsenoside Re, ginsenoside Ro, ginsenoside Rb1 and chikusetsu saponin Ⅳa in rat plasma and the pharmacokinetic parame- ters were evaluated by DAS 2.1.1 software. Results The main pharmacokinetic parameters of ginsenoside Re, ginsenoside Ro, ginsenoside Rbl and chikusetsu saponin Ⅳa were as follows: t1/2 (13.72 ± 9.14), (6.94 ± 3.51), (28.80±9.99), and (5.17±2.41) h; tmax(1 ±0.00), (4±0.00), (2±0.00), and (4±0.00) h; Cmax (370.27± 88.27), (649.06±98.22), (111.10±3.38), and (686.94±145.55) ng mL-1; AUC0-t (2188.75±599.81), (4743.44±509.26), (1699.06±35.66), and (4427.82± 1396.29) μg h L-1. Conclusion HPLC-MS and the pharmacokinetic parameters can be used to determine the pharmacokinetics of Tengzhu Weikang granules.
