草果挥发油作促渗剂的磷酸川芎嗪贴剂对大鼠心肌缺血的影响 被引量:6

Effects of Ligustrazine phosphate patches with the volatile oil of Tsaoko Fructus as penetration enhancers on myocardial ischemia in rats

摘要 目的观察3%草果挥发油作促渗剂的磷酸川芎嗪贴剂对大鼠急性心肌缺血的保护作用及凝血时间的影响。方法采用舌下静脉注射垂体后叶素(Pit)诱导大鼠急性心肌缺血模型。将大鼠随机分为模型组、基质组、赋形剂组、阳性对照(ig磷酸川芎嗪)组和1、2、4 cm^2磷酸川芎嗪贴剂组,观察各组大鼠注射Pit后不同时间点II导联心电图S-T段偏移和T波变化的情况,用毛细血管法测定大鼠的凝血时间。结果 1、2、4 cm^2磷酸川芎嗪贴剂均不同程度地抑制了Pit所致大鼠15~600 s内心电图S-T段和T波的变化,且能明显延长大鼠的凝血时间。结论含3%草果挥发油的磷酸川芎嗪贴剂对大鼠急性心肌缺血有保护作用且具抗凝血作用。 OBJECTIVE To investigate the protective effects of Ligustrazine phosphate (LP) patches with 3% Tsaoko Fructus volatile oil as penetration enhancers on myocardial ischemia rats and the effect on blood clotting time of rats. METHODS The animal model of myocardial ischemia induced by injecting Pituitrin (Pit) through sublingual vein was built to evaluate the efficacy of LP patches against myocardial ischemia in rats. Rats were randomly divided into 7 groups involving model group, matrix group, excipient group, positive control( ig LP solution) group, patches of 1 cm2 dose group, patches of 2 cmz dose group and patches of 4 cm2 dose group. The migration of S -T period and abnormality of T wave at different time points of standard Ⅱ lead electrocardiogram(ECG) in rats of myocardial ischemia were observed. The blood clotting time was surveyed by the capillary method. RESULTS The displace- ments of S -T period and T wave in 15 -600 s of ECG could be improved obviously on acute myocardial ischemia rats caused by Pit, and the blood clotting time could be extended after 3 doses ( 1, 2, 4 cm2 ) of LP patches were administrated. CONCLUSION LP patches with 3% Tsaoko Fructus volatile oil can protect the myocardial ischemia rats obviously and has the anticoagulation activity.
