HPLC测定降脂酮Ⅱ号中的5,7,4'-三甲基染料木素及其有关物质

Determination of 5,7,4'-trimethyl genistein and the related substance in JiangzhitongⅡ by HPLC

目的采用HPLC法测定降脂酮Ⅱ号中5,7,4'-三甲基染料木素(TMG)的含量及其有关物质。方法色谱柱为Kromasil C_(18)柱(150 mm×4.6 mm,5μm),柱温30℃,流动相为甲醇-水(80∶20),流速1 m L·min^(-1),检测波长260 nm。结果0.024~0.24μg TMG与峰面积具良好的线性关系(r=0.9999),平均回收率为99.42%,RSD=1.00%(n=6)。结论所用方法快速、简便、准确、专属性强,可用于降脂酮Ⅱ号的质量控制。

OBJECTIVE To establish an HPLC method to determine the content of 5,7,4＇ - trimethyl genistin （TMG） and the related substances in Jiangzhitong Ⅱ. METHODS Kromasil C18 column （ 150 mm ×4.6 mm, 5 μm） was used with a mobile phase of methanol - water （ 80 ： 20 ） and the flow rate of 1.0 mL. min - 1. The detection wavelength was 260 nm and the temperature was 30 ℃. RESULTS TMG presented a good linear correlation in the range of 0. 024 - 0. 240 μg （ r = 0. 9999 ）. The average recovery was 99.42% with the RSD of 1.00%. CONCLUSION This method is rapid, simple, specific and accurate. It can be used for the quality control of Jiangzhitong Ⅱ.
