浸取剂-超声波协同提取苦瓜皂苷的工艺优化

摘要 以乙醇、乙酸乙酯、环己烷及其两两混合作为浸取剂,利用超声波辅助提取苦瓜皂苷,考察了不同浸提液在不同超声功率、提取温度、料液比及提取时间下对苦瓜总皂苷得率的影响。以乙醇-乙酸乙酯作浸提剂时苦瓜皂苷总得率最高。在单因素实验的基础上,利用Box-Behnken设计四因素三水平响应面试验,确定最佳提取工艺条件为:超声功率350 W、提取温度48℃、料液比1∶25、提取时间3.0 h,在此工艺条件下,总皂苷得率为3.47%。 The momordicoside was extracted assisted by ultrasonic in leaching system using ethanol, acetidine, cyclohexane and the mixture of two above solvents as leaching agents. The effects of ultrasonic power, extraction temperature, mass ratio of raw material to solvent and extraction time on momordicoside yield were studied. The highest yield was obtained in ethanol-acetidine system. A Box-Behnken design involving four factors at three levels each was used to determine the optimum extraction conditions based on the single factor testing. The optimum extraction conditions were obtained as follows : ultrasonic power was 350 W, extraction temperature was 48 ℃, raw material to solvent ratio was 1 : 25, and extraction time was 3.0 h, under these conditions, the yield of momordicoside was 3.47%.
