促神经因子保护剂N6对脊髓损伤大鼠模型疗效的观察研究

摘要 目的通过大鼠的脊髓损伤模型,观察和探讨促神经因子保护剂N6对于脊髓损伤后运动功能的恢复作用和临床疗效,以期为进一步的研究提供思路和相关理论基础。方法 2017年3月将40只雄性SD大鼠平均随机分为4组,每组为10只。按照治疗方法的不同,分别分为:鸡尾酒促神经因子保护剂N6给药组(Cocktail组),甲强龙组(MP组),假手术组(Sham组)和单纯脊髓损伤组(SCI组)。前两组在建立大鼠脊髓损伤模型后10min即给予相应的药物。在建模及之后每隔7d观察和记录不同组大鼠的后肢功能运动评分(Basso-Beattie-Bresnahan,BBB评分)。结果 Sham组的大鼠BBB评分在建模后始终高于其余3组。Cocktail组和MP组在建模7dBBB评分高于SCI组(P<0.05),但Cocktail组与MP组相比差异无统计学意义(P>0.05)。在建模14、21、28d,Cocktail组的BBB评分高于其他2组(P<0.05)。结论使用鸡尾酒神经保护剂N6进行脊髓损伤的治疗,有利于大鼠运动功能的恢复。在脊髓损伤的临床研究和治疗中可以进一步进行探索和推广。 Objective To understand the recovery mechanisms and the clinic effect of N6 after spinal cord injury,so as to provide ideas and related theories for further research. Methods A total of 40 SD rats were randomly divided into 4 groups with different methods ;intrathecal injection of cocktail neuroprotectant N6 group(Cocktail) ;intrathecal injection of methylprednisolone group(MP);sham group(Sham)and spinal cord injury group(SCI). The rat model of acute SCI was established for the first 2 groups, 10min later, treatment groups were given corresponding drug treatment. Basso-Beattie- Bresnahan(BBB) score testing was employed to evaluate the remaining rats" locomotor function of hindlimbs on 1,7,14, 21,28 d after operation. Results The BBB score in Sham was significantly higher than that of the other 3 groups from beginning to end. Compared with SCI,BBB of cocktail and MP group was higher( P 〉0.05), however, there was no sig- nificant difference on 7th day between Cocktail and MP( P 〉0.05). The BBB score of group Cocktail was significantly higher on 14d,21d and 28 d( P〈0. 05). Conclusion Cocktail neuroproteetant N6 can promote the recovery of hindlimb motor function of SCI rats,which can be further studied and promoted in clinic.
