摘要
目的:探讨MDM2拮抗剂Nutlin-3促进肝癌细胞SMMC-7721发生焦亡的作用及相关机制。方法:Western blot检测细胞中活化caspase-1(p20)及IL-1β的表达,LDH法检测SMMC-7721细胞焦亡情况,ELISA检测细胞上清中IL-1β释放情况。结果:Nutlin-3提高SMMC-7721细胞活化caspase-1(p20)及IL-1β的蛋白表达水平。Nutlin-3处理显著提高了SMMC-7721细胞培养上清中的LDH及IL-1β的表达水平(P<0.05)。结论:Nutlin-3激活了caspase-1,诱导肝癌细胞SMMC-7721发生焦亡,并促进IL-1β释放。
Objective :To evaluate the effect of Nutlin-3, a MDM2 antagonist, on the pyroptosis in SMMC-7721 cells. Methods: The expression of actived caspase-1 ( p20 ) and IL-1 β was detected using Western blot analysis. Pyroptosis was investigated by a standard lactate dehydmgenase release assay(LDH). IL-1 β content in the cell culture supernatant was quantified by ELISA. Results: Nutlin-3 up-regulated the expression level of actived caspase-1 and IL-I[3 in SMMC-7721 cells. Meanwhile, Nutlin-3 increased significantly the content of LDH and IL-1 β in the cell culture supernatant ( P〈0.05 ). Conclusion : Nutlin-3 activated pro-caspase-1, promoted pyroptosis and IL-1β release in SMMC-7721 cells.
