Induction of Differentiation of Human Acute Myeloid Leukemia HL-60 Cells by 5-ally-7-gen-difluormethylenechrysin

摘要 目的:研究5-烯丙基-7-二氟甲基白杨素(ADFMChR)诱导人急性髓性白血病HL-60细胞分化作用。方法:体外培养人急性髓性白血病HL-60细胞。瑞氏-吉姆萨染色法观察细胞分化的形态学改变;细胞免疫组织化学法和间接免疫荧光显微镜观察细胞表面分化抗原CD11b表达。结果:瑞氏-吉姆萨染色结果显示,ADFMChR诱导HL-60细胞发生分化成熟的形态学变化;细胞免疫组织化学显示ADFMChR增加HL-60细胞粒细胞系的特异性表面分化抗原CD11b的表达;间接免疫荧光显示经ADFMChR处理的HL-60细胞表面分化抗原CD11b表达增强,阳性表达率呈剂量依赖性升高。结论:ADFMChR具有诱导HL-60细胞分化的作用。 Objective To investigate the effects of 5-ally-7-gen-difluormethylene- chrysin on differentiation of human acute myeloid leukemia HL-60 cell line cultured in vitro. Methods HL-60 cells were cultured in vitro. The morphology change of cell differentiation was observed by Wright-Giemsa stain. The expression of cell surface differentiation antigen CDllb of HL-60 was also observed using the cell immunohistochemical method and indirect fluorescence microscope. Results Results by Wright-Giemsa stain display that the cell morphology was changed toward maturity differentiation after treatment with ADFMChR. ADFMChR has the effect to induce HL-60 differenti-ation towards granulocyte series. The cell membranaceous surface differentiation antigen CDllb was expressed and its positive express rate showed increase in a dose-dependent manner. Conclusion ADFMChR has the ability to induce HL-60 differentiation.
