摘要
目的:观察紫花牡荆素(Casticin,CAS)诱导人宫颈癌HeLa细胞凋亡作用。方法:体外培养HeLa细胞。ELISA法测定细胞培养液组蛋白/DNA碎片。碘化丙啶(PI)染色流式细胞术(FCM)分析Sub-G1细胞百分率。ELISA法检测细胞Caspase-3活性。结果:CAS以浓度依赖方式增加HeLa细胞组蛋白/DNA碎片的渗漏(P<0.05),同时显著升高HeLa细胞Sub-G1百分率(P<0.05)。CAS以Caspase依赖方式激活HeLa细胞Caspase-3(P<0.05)。结论:CAS能有效诱导HeLa细胞凋亡。
Objective To investigate the effects of casticin(CAS) on apoptosis of human cervical cancer HeLa cell line.Methods Human cervical cancer HeLa cells were cultured in vitro.Histone/DNA fragments in medium were determined using ELISA assay.The percentage of the sub-G1 population was determined by flow cytometry of PI fluorescence staining.Caspase-3 activity in HeLa cells was examined using ELISA assay.Results CAS significantly increased leakage of the histone/DNA fragments in HeLa cell line(P<0.05),in a concentration-dependent manner.Meanwhile,CAS significantly promoted the increase of the percentage of the sub-G1 cell population in human cervical cancer HeLa cells(P<0.05).CAS effectively activated Caspase-3 in human cervical cancer HeLa cells(P <0.05),in a concentration-dependent manner.Conclusion CAS can effectively induce apoptosis of human cervical cancer HeLa cells.
