摘要 目的:研究5,7-二甲氧基黄酮(5,7-dimethoxyflavone,DMF)诱导人前单核细胞系U937细胞凋亡作用。方法:体外培养U937细胞,PI染色FCM分析Sub-G1细胞百分率;ELISA法检测细胞培养液组蛋白/DNA碎片、Caspase-9和Caspase-3活性。结果:DMF增高U937细胞Sub-G1百分率(P<0.05),呈浓度依赖性。DMF以浓度依赖方式刺激U937细胞组蛋白/DNA碎片渗漏(P<0.05)。DMF能有效诱导U937细胞Caspase-9和Caspase-3活化。结论:DMF诱导U937细胞凋亡作用与其激活线粒体途径相关。 Objective To investigate the effects of 5,7-dimethoxyflavone(DMF) on induction of apoptosis in U937 human promonocytic cells.Methods U937 human promonocytic cells were cultured in vitro.The percentage of sub-G1 cell population was determined by flow cytometry using PI fluorescence staining.Histone/ DNA fragment,Caspase-9 and Caspase-3 activity was determined using ELISA assay.Results DMF significantly increased the percentage of sub-G1 cell population in U937 cells,in a concentration-dependent manner(P<0.05).DMF promoted the leakage of the histone/DNA fragments in U937 cells(P<0.05),in a concentration-dependent manner.DMF can effectively activated Caspase-9 and Caspase-3 activity in in U937 cells.Conclusion DMF-induced apoptosis of U937 human promonocytic cells is associated with activation of mitochondrial apoptotic pathway.
