芹菜素选择性抑制人胃癌细胞活力和诱导细胞凋亡 被引量:3

Selectively Inhibition of Cell Viability and Induction of Apoptosis in Human Gastric Cancer BGC-823 Cell Line by Apigenin

摘要 目的:对比研究芹菜素对人胃癌BGC-823细胞和永生化人胃粘膜上皮GES-1细胞活力和细胞凋亡的影响。方法:体外培养BGC-823和GES-1细胞。MTS比色法测定细胞活力。PI染色FCM分析细胞凋亡率。ELISA法检测细胞Caspase-3活性。结果:芹菜素对BGC-823细胞活力有显著抑制作用(P<0.05),呈浓度依赖性,其IC50是22μmol/L;然而,对GES-1细胞活力抑制作用弱,其IC50达到77μmol/L;选择指数是3.5(77/22)。芹菜素选择性诱导BGC-823细胞Sub G1百分率增高(P<0.05),同时,活化BGC-823细胞Caspase-3(P<0.05)。结论:芹菜素具有选择性抑制人胃癌BGC-823细胞活力和诱导细胞凋亡作用。 Objective To investigate whether apigenin(API) can selectively inhibit cell viability and induce apoptosis in human gastric cancer BGC-823 cell line.Methods Human gastric cancer BGC-823 cells and immortalized human gastric mucosa epithelial GES-1 cells were cultured in vitro.Cell viability was determined using MTS colorimetric assay.The percentage of sub-G1 cell population was examined by flow cytometry(FCM) of PI fluorescence staining.Caspase-3 activity was determined using ELISA assay.Results Apigenin significantly inhibited the viability of BGC-823 cells(P<0.05),in a concentration-dependent manner,with IC50 is 22 μmol/L,had little effect on GES-1 cells,with IC50 is 77 μmol/L.Selective index of apigenin to BGC-823 cells is 3.5(77/ 22).Apigenin selectively increased the percentage of sub-G1 cell population(P<0.05),meanwhile,activatived Caspase-3 activity in human gastric cancer BGC-823 cells.Conclusion Apigenin selectively inhibited cell viability and induced apoptosis in human gastric cancer BGC-823 cell line.
