摘要
目的:观察不同浓度脂多糖(LPS)刺激人支气管上皮细胞(HBECs)不同时间后β防御素3基因(hBD-3Mrna))的表达,探索hBD-3在呼吸道细菌感染中的作用及其时效关系。方法:对照组和不同浓度LPS组(0.01、0.1、1、10μg/mL)诱导HBECs后,分别在不同时间点(1h、2h、4h、8h)行RT-PCR法检测hBD-3mRNA的表达。结果:hBD-3mRNA在对照组人支气管上皮细胞中有微量表达,不同时间组间无显著性差异(P>0.05);hBD-3mRNA在不同浓度LPS组中,随刺激时间增加hBD-3mRNA表达亦随之增加,不同时间组间比较有显著性差异(P<0.01)。结论:LPS能诱导HBECshBD-3表达上调,并在一定范围内呈时间依赖性,hBD-3可能参与气道对LPS最初的防御反应。
Objective To observe the mRNA expression at a series of time of human β-defensin-3(hBD-3) induced by lipopolysaccharide(LPS) of different concentration in human bronchial epithelial(HBE) cells,and to explore the results ts and time-effect to respiration track infection.Methods Human bronchial epithelial cells were stimulated with control-group and different concentration LPS group(0.01、0.1、1、10μg/mL) and the mRNA expression of hBD-3 at different time(1h、2h、4h、8h) was detected by RT-PCR.Results A microscale expression of hBD-3 mRNA was found in control group and significant difference had not found in different time-group(P>0.05).After stimulated by different concentration LPS,the expression of hBD-3 mRNA in normal human bronchial epithelial cells increased with the time increased;and there were significantly difference in different time-group(P<0.01).Conclusion After being stimulated by LPS,the expression of hBD-3 was increased.LPS can induce the expression of hBD-3 in a time dependent manner at a certain range.The expression of hBD-3 may play an initial defensive role against bacterial invasion.
