摘要
目的:探讨自噬基因beclin-1在儿童急性淋巴细胞白血病(ALL)患者外周血白细胞的表达及意义。方法:选取儿童急性淋巴细胞白血病患者39例,体检正常儿童40例作为对照,收集患者及正常对照外周血标本,分离白细胞,用免疫组化和免疫印迹法检测beclin-1表达,分析比较患者与对照组beclin-1蛋白表达差异。结果:ALL患儿外周血白细胞beclin-1表达阳性率48.7%(19/39),低于正常对照组82.5%(33/40)(P<0.005),Western blot显示患儿阳性标本表达量亦低于对照阳性标本。标危组表达率(59.3%)高于高危组(25.0%),B-ALL与T-ALL的beclin-1表达率差异无显著性(P<0.05)。结论:beclin-1表达率及表达量在ALL患儿外周血白细胞中明显低于正常对照;beclin-1表达率与诊断时免疫学分型无明显关系,标危组表达率高于高危组。beclin-1可能与儿童ALL发生发展相关。
Objective To explore the role of autophage related protein beclin-1 in the pathogenesis of acute lymphyblastic leukemia(ALL) by investigating the expression of beclin-1 in peripheral leucocytes of children patients with ALL.Methods Peripheral leucocytes from 39 children patients with ALL and 40 normal childern as control were examined for beclin-1 expression using immunohistochemistry staining and Western blot.Analysis the diffenrence of beclin-1 expression rates and levels between the ALL patients and normal children.Results The beclin-1 expression rates of ALL children patients and control group were 48.7%(19/39) and 82.5%(31/40)respectively.There are significantly differences between the two groups(χ2=10.017,P=0.002).The beclin-1 expression level in ALL patients was lower than that of control group.No statistic differences were found between B-ALL and T-ALL(χ2=0.300,P=0.584).The ALL patients of low risk group are more likely to have a high beclin-1 expression compared with those of high risk group(χ2=3.903,P=0.046).Conclusion The beclin-1 expression rate and levels were down-regualted significantly in peripheral leucocytes of ALL patients.It suggests that beclin-1 may be involved in the pathagenesis mechanism of ALL.
