葡萄籽提取物原花青素对大鼠实验性隐睾生精细胞凋亡的影响 被引量:2

Effect of Grape Seed Proanthocyanidin Extract on Spermatogenic Cell Apoptosis in Rats Experimental Cryptorchidism

摘要 目的:探讨葡萄籽提取物原花青素对隐睾生精细胞凋亡的影响,为临床治疗某些特发性男性不育症药物的开发研究提供实验依据。方法:将雄性健康SD大鼠40只随机分为隐睾+原花青素(高、中、低剂量)组、隐睾+生理盐水组、假手术组5组,每组8只大鼠,每天定时定量灌胃3次,7天后断颈处死大鼠,双侧睾丸称湿重并取手术侧睾丸HE染色观察组织学变化,TUNEL法检测细胞凋亡。结果:与生理盐水组比较,原花青素各剂量组的生精细胞凋亡程度显著减轻(P<0.05),且呈剂量效应。结论:原花青素能够减轻雄性SD大鼠实验性隐睾生精细胞凋亡的程度。 Objective To study the biological effects of grape seed proanthocyanidin extract(GSPE) on spermatogenesis cell apoptosis with the purpose of providing experiment proofs of the pharmacological development study for the clinical treatment of the idiopathic male infertility.Methods Forty immature male Sprague-Dawley rats(42 day old) were divided into five groups: crypt orchid+ GSPE(high,medium,low),crypt orchid +natural salt,sham operation.Each group has 8 rats.The rats of crypt orchid+ GSPE(high,medium,low) group was administered intragastrically with GSPE accordingly.The rats were Killed by cervical on the 7th day after operation.Lateral test ides were dissected and weighted,undergo test ides were performed HE staining to observe histological character(tissue histology) by TUNEL method.Results Compared to the crypt orchid + normal saline group,the degree of apoptosis of spermatogenic cells treated by orchid+ GSPE(high,medium,low) significantly reduced(P <0.05)and it exhibits dose dependent effect.Conclusion GSPE may inhibit the spermatogenic cell apoptosis of the immature male Sprague-Dawley rats.
