摘要
目的:探讨h1 CaP(h1 calponin,碱性调宁蛋白)及ERK1/2(external signal regulated kinase1/2,细胞外信号调节激酶1/2)在妊娠小鼠子宫平滑肌细胞中总蛋白和磷酸化蛋白的表达变化及与分娩的关系。方法:按妊娠阶段的不同,将小鼠分为未孕组、孕7天组、孕12天组、孕17天组、孕18天组、孕19天组、产时组。应用Western blot检测子宫平滑肌中h1 CaP、磷酸化h1 CaP(ph1 CaP)、ERK1/2、磷酸化ERK1/2(p ERK1/2)的蛋白表达。结果:产时组h1 CaP蛋白表达显著高于未孕、孕7天、孕12天、孕17天组(P<0.01),与孕18天、孕19天组比较差异无统计学意义(P>0.05);ERK1总蛋白表达量产时组与其他各组比较差异无统计学意义(P>0.05);ERK2总蛋白表达量产时组与其他各组比较差异无统计学意义(P>0.05)。ph1 CaP/h1 CaP的比值为产时组显著高于未孕组、孕7天、孕12天、孕17天、孕18天、孕19天组(P<0.01),;pERK1/ERK1的比值产时组与其他各组比较差异无统计学意义(P>0.05);pERK2/ERK2的比值产时组显著高于未孕组、孕7天、孕12天、孕17天、孕18天、孕19天组(P<0.01)。结论:子宫平滑肌细胞中h1 CaP蛋白表达在临产前达高峰,ph1 CaP及p ERK2水平在临产时达最高峰。h1 CaP可通过促进ERK2磷酸化参与分娩发动;h1 CaP对分娩发动的影响不通过ERK1途径。
Objective To evaluate the expression and phosphorylation of h1 calponin(h1 CaP) and external signal regulated kinase1/2(ERK1/2) in pregnant mice myometrium during pregnancy and labor.Methods The pregnant mice were divided into severn groups: non-pregnancy(NP),gestational day 7(D7),gestational day 12(D12),gestational day17(D17),gestational day18(D18),gestational day 19(D19) and in labour(IL) groups.The protein expression was detected by Western blot in mice myometrium.Results H1 Cap protein level in IL group was significantly higher than that in NP,D7,D12,D17 groups(P<0.01),but not significantly different than that in D18 and D19 groups(P>0.05).There was no significantly difference in ERK1 and ERK2 protein level among IL group and NP,D7,D12,D17,D18 and D19 groups(P>0.05).The ph1CaP/h1CaP ratio in IL group was significantly higher than that in NP,D7,D12,D17,D18 and D19 groups(P<0.01).There was no significantly difference in pERK1/ERK1 ratio among IL group and NP,D7,D12,D17,D18 and D19 groups(P>0.05).The pERK2/ERK2 ratio in IL group was significantly higher than that in NP,D7,D12,D17,D18 and D19 groups(P<0.01).Conclusion The expression of h1CaP in mice myometrium reach the peak at late pregnancy.The phosphorylation of h1CaP and ERK2 both reach the peak at parturition.Phosphorylation of ERK2 mediated by the H1Cap is suggested to contribute to the initiation of labour.Phosphorylation of ERK1 may be not mediated by the h1Cap during labour.
